Summary: This study reveals that surgery leads to neuronal hypoactivity and impairs learning-dependent dendritic spine formation, resulting in deficits in multiple learning tasks. These neuronal and synaptic alterations are mediated by peripheral monocytes through NLRP3 inflammasome-dependent IL-1β production. Decreasing peripheral monocytes or inactivating NLRP3 inflammasomes can reduce IL-1β levels and improve neuronal and behavioral deficits.

Summary: This study reveals that microglia activate NLRP3 inflammasomes in neurons and endothelial cells via IL-1 beta/IL-1R1/TRAF6 signaling pathway, promoting cerebral ischemia and reperfusion injury. Inhibition of IL-1R1 can block inflammatory damage and prevent excessive infiltration of leukocytes into the brain, leading to better neurological outcome.

Summary: The depletion of NAD(+) affects the innate immune potential of macrophages and has an impact on NLRP3 activation. FK866 facilitates NLRP3 activation by promoting perinuclear clustering of mitochondria, while supplementation with NAD(+) can counteract the FK866-induced caspase-1 cleavage.

Summary: The phosphorylation of NLRP3 was shown to be a crucial step for inflammasome priming in macrophages, with Ser725 being phosphorylated directly by MINK1 kinase. MINK1 deficiency reduced NLRP3 activation and inflammatory responses. Additionally, ROS were found to upregulate MINK1 activity and promote inflammasome priming through Ser725 phosphorylation, suggesting a potential intervention strategy for inflammasome-related diseases.

Summary: Chen et al. found that spreading depolarization activates the neuronal NLRP3 inflammasome and its downstream inflammatory cascades, causing cortical neuroinflammation and trigeminovascular activation. The activation of NLRP3 inflammasome is exclusive to neurons and is facilitated by microglial activation in the context of multiple spreading depolarizations.

Summary: Bacterial infections can activate hematopoietic stem and progenitor cells to migrate from the bone marrow to the spleen, a process known as extramedullary hematopoiesis. Recent studies have shown that gut commensal bacteria not only regulate the host immune system, but also hematopoietic homeostasis. This study reveals that Akkermansia muciniphila, a mucin-degrading bacterium, can rapidly activate bone marrow myelopoiesis and induce delayed extramedullary hematopoiesis through the MYD88/TRIF innate immune signaling pathway.

Summary: This study demonstrates that the scaffold protein RACK1 plays a crucial role in promoting NLRP3 inflammasome activation induced by Pasteurella multocida. RACK1 downregulation reduces cell death and bacterial invasion, while RACK1 overexpression enhances inflammasome activation. Furthermore, RACK1 regulates NF-kappa B and promotes inflammasome assembly to facilitate NLRP3 inflammasome activation.

Summary: Cells face continuous challenges from tissue insult caused by endogenous and exogenous stimuli. Innate immunity, especially through Toll-like receptors and NOD-like receptors, plays a crucial role in recognizing danger signals and activating the inflammasome complex to initiate pyroptosis. This review discusses the molecular events involved in these processes and explores experimental NLRP3 inhibitors.

Summary: Pyroptosis is a highly inflammatory form of programmed cell death triggered by pro-inflammatory signals, characterized by cell swelling and release of cytokines. In LPS-induced inflammation, the mechanism of endometrial epithelial cell pyroptosis remains unclear, possibly involving specific inflammasome cascade pathways.

Summary: The newly discovered cITLN is a potential player in intestinal mucosal immunity, able to agglutinate both gram-positive and gram-negative bacteria with a stronger effect on the former. This differs from previous expectations as it was not found to inhibit bacterial growth but rather enhance bacterial agglutination through binding to peptidoglycan (PGN).

Summary: In this study, transcriptome analysis was performed to investigate the immune response of common carp against SVCV infection. It was found that the RLR signaling pathway was significantly enriched after SVCV infection. Further investigation revealed that CcRIG-I and CcTRIM25 suppressed the replication of SVCV by inducing the phosphorylation of TBK1, IRF3, and p65, as well as the expression of ifn-1, viperin, isg15, and mx. Additionally, CcTRIM25 was shown to regulate CcRIG-I-mediated immune response through interaction with CARD and promoting K63-linked polyubiquitination of CcRIG-I.

Summary: Mammalian cells activate HIFs to survive in low-oxygen environments, but injury-induced hypoxia is unable to activate HIF1 alpha. However, IL-17A derived from ROR gamma t(+) gamma delta T cells can activate HIF1 alpha. The IL-17A-HIF1 alpha axis promotes glycolysis for wound healing in epithelial cells, and loss of IL-17RC, HIF1 alpha, or blockade of glycolysis disrupts the repair process. These findings highlight the interconnectedness of inflammation, metabolism, and migration programs, as well as the role of immune cell-derived inputs in cellular adaptation to hypoxic stress during repair.
