Cutting Edge: TRAF6 Mediates TLR/IL-1R Signaling-Induced Nontranscriptional Priming of the NLRP3 Inflammasome

NLRP3 inflammasome activiation requires two sequential signals. The priming signal 1 fromTLRs or cytokine receptors induces the transcription of NLRP3 and IL-1b, and concomitantly promotes transcription-independent activation of caspase-1. The activating signal 2 can be provided by microbial products or danger signals. In this study we found that TRAF6 is necessary for the non-transcriptional priming of NLRP3 inflammasome by TLR/IL-1R derived signals. Deficiency of TRAF6 specifically inhibited TLR/IL-1R priming-initiated caspase-1 cleavage, pyroptosis, and secretion of presynthesized IL-18. Mechanistically, TRAF6 promoted NLRP3 oligomerization as well as the interaction between NLRP3 and apoptosis-associated speck-like protein containing a CARD. Of note, the nontranscriptional priming via TRAF6 did not involve mitochondrial reactive oxygen species or the phosphorylation of Jnk, Erk, and Syk, whereas the ubiquitin E3 ligase activity of TRAF6 was required. Our findings thus extended cognition on the mechanism of NLRP3 inflammasome activation, and provided a novel target for controlling NLRP3-related diseases.

TRIM25 inhibits spring viraemia of carp virus replication by positively regulating RIG-I signaling pathway in common carp (Cyprinus carpio L.)

Summary: In this study, transcriptome analysis was performed to investigate the immune response of common carp against SVCV infection. It was found that the RLR signaling pathway was significantly enriched after SVCV infection. Further investigation revealed that CcRIG-I and CcTRIM25 suppressed the replication of SVCV by inducing the phosphorylation of TBK1, IRF3, and p65, as well as the expression of ifn-1, viperin, isg15, and mx. Additionally, CcTRIM25 was shown to regulate CcRIG-I-mediated immune response through interaction with CARD and promoting K63-linked polyubiquitination of CcRIG-I.

Interleukin-17 governs hypoxic adaptation of injured epithelium

Summary: Mammalian cells activate HIFs to survive in low-oxygen environments, but injury-induced hypoxia is unable to activate HIF1 alpha. However, IL-17A derived from ROR gamma t(+) gamma delta T cells can activate HIF1 alpha. The IL-17A-HIF1 alpha axis promotes glycolysis for wound healing in epithelial cells, and loss of IL-17RC, HIF1 alpha, or blockade of glycolysis disrupts the repair process. These findings highlight the interconnectedness of inflammation, metabolism, and migration programs, as well as the role of immune cell-derived inputs in cellular adaptation to hypoxic stress during repair.

Negative regulation of interferon regulatory factor 6 (IRF6) in interferon and NF-?B signalling pathways of common carp (Cyprinus carpio L.)

Summary: This study cloned the CcIRF6 gene from common carp and investigated its role in the immune response of fish. The results showed that CcIRF6 is involved in the antiviral and antibacterial immune response and negatively regulates the expression of IFN and NF-kappa B signaling pathways. This study provides a theoretical basis for the study and prevention of fish disease pathogenesis.
