Summary: Berberine was extracted from Berberis vulgaris, Berberis aquifolium, and Hydrastis canadensis using ethanol and water (70:30, v/v). The extracted berberine was characterized using UV-visible and Fourier-transform infrared spectroscopy. Thin-layer chromatography and high-performance liquid chromatography (HPLC) methods were both employed for the separation and identification of berberine, and showed different applications. The extracted berberine exhibited better activity in treating Leishmaniosis compared to the standard drug Amphotericin B.

Summary: Bioanalytical profiling of ethyl acetate extracts from Salvia aegyptiaca, S. verbenaca, and S. officinalis showed the presence of five bioactive zones. The compounds present in these zones exhibited antibacterial, antifungal, radical scavenging, and enzyme inhibitory activities. Triterpenes, linoleic acid, and linolenic acid were identified as the bioactive compounds in these extracts.

Summary: Ambient desorption/ionization mass spectrometry (ADI-MS) is a rapid screening tool that allows the analysis of samples without sample preparation. However, quantitative analysis of analytes in complex matrices remains a challenge. In this study, a carefully selected surface and co-deposited isotopically labeled standards were found to significantly improve the capabilities of FAPA-HR-MS for direct quantitative analysis. The use of TLC-based surfaces as sample carriers was shown to be an attractive tool for fast and efficient mass spectrometric investigations of complex samples.

Summary: High-performance thin-layer chromatography is a promising technique for high-throughput separations. The sample preparation is crucial to control matrix interferences and improve the detectability of target compounds. Recent trends have shifted towards bioassays and mass spectrometry. Traditional methods are being replaced by automated instrumental approaches for faster and streamlined sample processing. Liquid-liquid extraction is commonly used, although solid-phase extraction is gaining popularity. Planar solid-phase extraction is increasingly used for complex samples. Factors driving current interest include faster and simpler sample workflows, analysis of more complex samples, and the determination of minor contaminants with high concentration factors.

Summary: A green, fast, high-sample-throughput, non-instrumental, and affordable analytical method based on surfactant-assisted dispersive liquid-liquid microextraction (SA-DLLME) combined with thin-layer chromatography-digital image colourimetry (TLC-DIC) was proposed for determining favipiravir in biological and pharmaceutical samples. The method showed good linear range, low LOD and LOQ, and close agreement with HPLC-UV analysis results. The proposed methodology is green in nature and can be applied in resource-limited laboratories during the COVID-19 pandemic.

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.
