Effects of prenatal alcohol consumption on cognitive development and ADHD-related behaviour in primary-school age: a multilevel study based on meconium ethyl glucuronide

BackgroundAlcohol intake during pregnancy is considered to be a risk factor for child development. Child biomarkers of intrauterine alcohol exposure have been rarely studied. We investigated whether a meconium alcohol metabolite (ethyl glucuronide, EtG) was associated with cognitive development, ADHD-related behaviour and neurophysiological markers of attention and executive control of children at primary-school age. MethodsMothers provided self-report on prenatal alcohol consumption during their 3rd trimester. Meconium samples were collected at birth. A total of 44 children with a meconium EtG above the detection limit (10ng/g) and 44 nonexposed matched controls were compared. A second threshold (154ng/g) was applied to study the dose effects. When children reached primary-school age, mothers rated ADHD-related behaviour, child cognitive development was measured using an IQ test battery, and event-related potentials were recorded during a cued go/nogo task. ResultsChildren in both EtG-positive groups allocated fewer attentional resources than controls to the go/nogo task (reduced P3 component in go-trials). Children with a meconium EtG above 154ng/g were also found to have an IQ that was six points lower than the other groups. Within the EtG154ng/g group, there was a positive correlation between EtG value and ADHD-related behaviour. These significant effects were not observed in relation to the maternal self-report data. ConclusionsAssociations between EtG and cognitive deficits, attentional resource capacity and ADHD-related behaviour could be documented with effects that were partially dose-dependent. In addition to maternal self-reports, this biomarker of intrauterine alcohol exposure may be considered as a predictor of child development.

Summary: Evidence from the BRIDGES study suggests that germline protein truncating variants (PTVs) in FANCM are associated with increased risk of ER-negative and triple-negative breast cancer (TNBC), particularly for those with a family history. This study further investigates the association between FANCM missense variants (MVs) and breast cancer risk using the BRIDGES study, analyzing a total of 689 MVs. The results indicate that FANCM MVs may be low/moderate risk factors for ER-negative and TNBC subtypes of breast cancer.

Summary: There is a lack of empirical research on non-pharmacological interventions for Alzheimer's disease (AD) globally. This paper examines the biochemical, clinical, and social aspects of AD in relation to psychological interventions and their impact on patients' quality of life. Despite limited research, psychological interventions based on biochemical and other studies show potential for improving emotional well-being and slowing disease progression in AD patients. However, no effective methods of AD treatment have been established to date.

Summary: This study identified 14 genes associated with breast cancer using gene-based aggregation analysis, including two newly discovered genes FMNL3 and AC058822.1. Furthermore, associations with established candidate genes like ESR1 were found through the collaboration of multi-ancestral cohorts, highlighting the importance of diversifying study cohorts. These findings provide new insights into the development of breast cancer.

Summary: The study evaluated the prognosis of patients with triple-negative breast cancer (TNBC) and other subtypes by using a biomarker panel including cytokeratin 5 (CK5), cluster of differentiation 117 (CD117), and epidermal growth factor receptor (EGFR). The results showed that the non-TBN subgroup of TNBC lesions with at least one positive biomarker (CK5, CD117, and/or EGFR) had longer disease-free survival (DFS) and overall survival (OS). This highlights the importance of molecular-based subclassifications of breast cancer for treatment options and prognosis stratification.

Development of Comorbid Depression after Social Fear Conditioning in Mice and Its Effects on Brain Sphingolipid Metabolism

Summary: Currently, there are no animal models available for studying specific social fear and social fear with comorbidities. This study investigated whether social fear conditioning (SFC), an animal model for social anxiety disorder (SAD), leads to comorbidities and how it affects brain sphingolipid metabolism. The results showed that SFC altered emotional behavior and brain sphingolipid metabolism in a time-dependent manner. Social fear alone did not cause changes in non-social anxiety or depressive behavior, but comorbid depression developed five weeks after SFC, accompanied by alterations in sphingolipid metabolism in various brain regions.

This study aimed to evaluate women's recall of prenatal events and found that retrospective self-reporting was not always accurate. The reliability of self-reporting on prenatal alcohol, smoking, partnership quality, pregnancy satisfaction, and obstetric complications varied.

Heart Rate as a Correlate for the Emotional Processing of Body Stimuli in Anorexia Nervosa

Summary: The study investigated aberrant emotional reactions toward body stimuli in anorexia nervosa patients and hypothesized altered heart rate reactions toward those highly significant and emotional stimuli. It found that anorexia nervosa patients showed greater overall change in heart rate relative to baseline when viewing pictures of women of different weight categories. Significant heart rate decelerations were observed for pictures of extremely underweight and overweight women, which were correlated with sociocultural attitudes toward appearance in the extremely underweight category.

Summary: This study analyzed paternal contributions to child well-being and found that father psychopathology significantly predicted girls' quality of life, while maternal parenting behavior had a significant influence on boys' quality of life. Fathers face different individual stressors and have different parenting behavior compared to mothers, but their influences are still significant, especially for daughters. Therefore, father-addressed pre- and intervention programs in child psychotherapeutic treatment are of high relevance.
