Four new phenolics and antiparasitic secondary metabolites from Flacourtia rukam Zoll. & Mortizi

Some compounds exhibited good anti-inflammatory and anti-Trypanosoma brucei activity, while all tested compounds were inactive against antileishmanial or antimalarial.

Application of a Biocatalytic Strategy for the Preparation of Tiancimycin-Based Antibody-Drug Conjugates Revealing Key Insights into Structure-Activity Relationships

Summary: Antibody-drug conjugates (ADCs) are cancer chemotherapeutics that combine an antibody-based delivery system, cytotoxic payload, and a chemical linker. Functionalization of the payload must be carefully designed to maintain the potency needed for ADC efficacy. In this study, we successfully translated biocatalytically functionalized TNMs into ADCs using a dual-variable domain (DVD)-mAb platform. We evaluated different linker chemistries and found a trade-off between potency and antigen selectivity. This work represents a novel approach in the development of ADCs for various cancer types.

The anti-infective potential of the endophytic fungi associated with Allium cepa supported by metabolomics analysis and docking studies

Summary: Endophytic fungi in Allium cepa were investigated for their potential as a source of anti-infective drugs. One isolate, Penicillium sp. LCEF10, showed significant anti-infective activity against several tested microbes and was identified as Penicillium oxalicum NRRL 787. Metabolomic analysis revealed the presence of various bioactive compounds in the ethyl acetate extract of Penicillium sp. LCEF10, including polyketides, macrolides, phenolics, and terpenoids. Molecular docking study predicted the compounds most likely responsible for the anti-infective activity.

Neogrisemycin, a Trisulfide-Bridged Angucycline, Produced upon Expressing the Thioangucycline Biosynthetic Gene Cluster in Streptomyces albus J1074

Summary: The authors isolated neogrisemycin (1) from a recombinant Streptomyces albus J1074 strain SB4061 expressing an engineered thioangucycline (TAC) biosynthetic gene cluster (BGC). They determined the structure and absolute configuration of 1 using mass spectrometry, nuclear magnetic resonance, and single-crystal X-ray diffraction analyses. They proposed that 1, like the TACs, is derived non-enzymatically from the common epoxide (8) encoded by the tac BGC, mediated by endogenous hydrogen trisulfide.

Multidrug-Resistant Acinetobacter baumannii Infections in the United Kingdom versus Egypt: Trends and Potential Natural Products Solutions

Summary: Acinetobacter baumannii is a global concern as it causes various infections, especially among immunocompromised patients in intensive care units. This pathogen has the ability to become resistant to almost all available antibiotics and spreads resistance at a high rate. Comparing the research efforts of the UK and Egypt, Egyptian researchers have made notable contributions in finding solutions through natural products. The incidence of A. baumannii infections has been increasing in Egypt, especially with alarming levels of antibiotic resistance. Studies in the UK have shown earlier adoption of high-throughput technologies for tracking and detecting A. baumannii and its resistance compared to Egypt.

Application of Ultraviolet-C Radiation and Gaseous Ozone for Microbial Inactivation on Different Materials

Summary: With the emergence of the COVID-19 pandemic, there has been a global push for environmentally sustainable and rapid sterilization methods. This study presents novel findings on the performance of combining ozone and ultraviolet-C radiation (UVC) for sterilization, which has shown improved effectiveness compared to their individual applications. The study also highlights the differing effects on fungi and bacteria when smeared or placed on nonabsorbent materials.

Aloin A inhibits SARS CoV-2 replication by targeting its binding with ACE2-Evidence from modeling-supported molecular dynamics simulation

Summary: The current study aimed to assess the inhibitory efficacy of aloin A against SARS CoV-2. In vitro testing showed weak to moderate activity against SARS CoV-2 proteases. However, aloin A efficiently inhibited the replication of SARS CoV-2 in cells. Further virtual screenings and simulations identified RBD-ACE2 as a key target for aloin A, suggesting its role as a destabilizer in the complex.

Summary: Neurological disorders can be treated by delivering Quercetin (QER) to the brain, and a study has developed transferosomal nanovesicles as a suitable nanocarrier for QER brain targeting. The optimized formulation exhibited sustained release and enhanced nasal mucosa permeability. The high stability and low cytotoxicity of the nanovesicles were confirmed through various experiments.
