Summary: A study developed a fast and simple single-step centrifugation protocol for producing high-quality platelet-free plasma for EV research. Compared to the traditional two-step protocol, the single-step protocol is faster, removes platelets effectively, and has a slightly higher plasma yield.

Summary: In this study, a comparative phosphoproteomic analysis of platelets from obese patients revealed up-regulated levels of phosphorylation events and altered signaling pathways compared to lean controls. Flow cytometry assays and mass spectrometry validation confirmed platelet dysfunction in obese individuals, including higher surface levels of GPVI and CLEC2. Soluble GPVI levels were also found to be elevated in plasma from obese patients, suggesting increased GPVI signaling activation.

Summary: The study demonstrated contrasting patterns in the intracellular ion concentrations of COAT-activated platelets compared to non-COAT platelets, with the former showing high and prolonged calcium concentration, transient sodium increase, and fast potassium efflux. Inhibition of the sodium-calcium exchanger (NCX) reduced calcium mobilization and prevented the formation of procoagulant platelets, highlighting the critical and dichotomous role of NCX during platelet activation induced by convulxin-plus-thrombin.

Summary: The aim of this study was to provide expert consensus guidance on the use of flow cytometry for the evaluation of platelet function in patients with low platelet counts. Through literature review and focus group discussions, consensus recommendations were made on sample collection, preanalytical variables, sample type, and results expression. These recommendations would allow standardization of protocols in future clinical studies.

Summary: Flow cytometry was used to analyze the immune cell composition in the peripheral blood and decidual tissue of women with recurrent pregnancy loss (RPL). The results showed abnormalities in the immune cell profiles of RPL patients, but further verification is needed to establish correlations with previous miscarriages.

Summary: The study aimed to develop a novel and practical protocol for micronucleus test in rat peripheral blood using flow cytometry, with method validation. The FCM-based method consistently demonstrated highly sensitive responses for micronucleus detection and correlated well with traditional Giemsa staining method. This method provides an efficient system for micronucleus scoring with easy operation and low background.

Summary: This study investigated the platelet pathophysiology in pediatric ECMO patients and found that the platelet response decreased over time, which may contribute to bleeding during ECMO. These findings are important for understanding the mechanisms of bleeding in pediatric ECMO and predicting or detecting bleeding and thrombosis.

Summary: Immunophenotyping in young C57BL/6J mice showed that the female reproductive cycle does not significantly impact blood immunophenotype, while sex-specific differences exist with fewer immune cells in female mice. Surprisingly, immunophenotype is more variable in male mice, with weight as a significant contributing factor.

Summary: Different biomarkers were investigated to detect the causes of severe complications in preterm infants. It was found that the counts of EVs in cord blood did not show significant differences between preterm and control groups, and there was no correlation with gestation age. Furthermore, samples with high levels of free hemoglobin had significantly higher counts of certain types of EVs. Further studies using more sensitive cytometers are needed to confirm the findings.

Summary: NGS has a superior diagnostic performance compared to CG in patients with suspected MDS, and mutations identified by NGS can alter patients' risk categories. Using an 'NGS-first approach' may significantly reduce the need for karyotyping.

Summary: By utilizing NGS as the primary genomic testing modality, a streamlined approach in testing suspected myeloid and plasma cell neoplasms patients was implemented, eliminating over three quarters of all CG requests and improving the financial case for adopting NGS. Key factors for the success of this study included collaboration between clinical and genomic diagnostic teams in developing the algorithm, rapid turnaround time for BM interpretation for triage, and communication between laboratories.

Summary: In a cost-effectiveness analysis comparing eltrombopag and IVIG for managing ITP in the perioperative setting, eltrombopag was found to be not only noninferior to IVIG but also less costly. The use of perioperative eltrombopag resulted in cost savings per patient, with the probability of it being cost-effective even without willingness to pay.
