Statistical analysis of molecule colocalization in bioimaging

The quantitative analysis of molecule interactions in bioimaging is key for understanding the molecular orchestration of cellular processes and is generally achieved through the study of the spatial colocalization between the different populations of molecules. Colocalization methods are traditionally divided into pixel-based methods that measure global correlation coefficients from the overlap between pixel intensities in different color channels, and object-based methods that first segment molecule spots and then analyze their spatial distributions with second-order statistics. Here, we present a review of such colocalization methods and give a quantitative comparison of their relative merits in different types of biological applications and contexts. We show on synthetic and biological images that object-based methods are more robust statistically than pixel-based methods, and allow moreover to quantify accurately the number of colocalized molecules. Cytokine receptor cluster size impacts its endocytosis and signaling

Three distinct populations of IL-2R gamma clusters were identified, with different sizes playing different roles in IL-2R internalization and signaling. Membrane cholesterol and the branched actin cytoskeleton were found to be key regulators of IL-2R gamma clustering and IL-2-induced signaling, affecting CIE initiation and signal transduction.

Summary: Secreted proteins are transported along intracellular route from the endoplasmic reticulum through the Golgi before reaching the plasma membrane. Small GTPase Rab and their effectors play a key role in membrane trafficking. MICAL-L1 is found to be associated with tubulo-vesicular structures and to have a significant colocalization with markers of the Golgi apparatus and recycling endosomes, revealing a new role in cargo delivery to the plasma membrane.

Imaging and Measuring Vesicular Acidification with a PlasmaMembrane-Targeted Ratiometric pH Probe

Summary: Tracking the pH variation of intracellular vesicles is crucial for assessing cell trafficking and metabolism. we designed a fluorogenic ratiometric pH probe, Mem-pH, which specifically targets the plasma membrane of cells and accumulates in intracellular vesicles through endocytosis. It allows for dynamic observation of vesicular acidification throughout the endocytic pathway and pH measurement through ratiometric imaging.

Tracking receptor motions at the plasma membrane reveals distinct effects of ligands on CCR5 dynamics depending on its dimerization status

Summary: This study investigated the impact of different states on the biological function and therapeutic targeting of CC chemokine receptor 5 (CCR5). The researchers used microscopy and statistical methods to track and classify the motion of different receptor subpopulations. The results showed that CCR5 exists in various ligand-free forms, stabilized by different ligands. Agonist stimulation restricted the mobility of CCR5 and led to its clustering, while inverse agonist stimulation had the opposite effect. The study also demonstrated the importance of CCR5 dimerization in its coupling to G proteins, immobilization and clustering upon activation, and receptor endocytosis.

Dual-Color Photoconvertible Fluorescent Probes Based on Directed Photooxidation Induced Conversion for Bioimaging

Summary: We introduce a new concept called Directed Photooxidation Induced Conversion (DPIC) to produce dual-color photoconvertible probes. Styryl-coumarins (SCs) with Aromatic Singlet Oxygen Reactive Moieties (ASORMs) have been synthesized to support this mechanism. SC-P, a yellow emitting probe with a pyrrole moiety, can convert to a stable blue emitting coumarin with a 68 nm shift, enabling photoconversion and tracking of lipid droplets in live cells. This novel approach may lead to a new generation of photoconvertible dyes for advanced bioimaging applications.

Impact of Green Gold Nanoparticle Coating on Internalization, Trafficking, and Efficiency for Photothermal Therapy of Skin Cancer

Summary: Skin cancer, including melanoma and nonmelanoma cancers, is a global health issue. This study focuses on basal cell carcinoma as a good prognosis skin cancer for the first clinical proof of concept. Photothermal therapy using gold nanoparticles synthesized by green chemistry (gGNPs) is investigated for its potential to treat skin cancers. The study demonstrates that gGNPs are less cytotoxic than gold nanoparticles synthesized by Turkevich's method and optimized gGNP coating and stabilization are described. Glucose-stabilized gGNPs (gGNP@G) show specific internalization in cancerous cells with no cytotoxicity, providing a promising approach for targeted cancer treatment.
