Compound 3 exhibited potent inhibitory activities against Staphylococcus aureus and methicillin-resistant S. aureus, as well as some weak antiviral activity against the SARS-CoV-2 main protease responsible for the COVID-19 pandemic.

Summary: Two new xanthones (1 and 2) were isolated from deep-sea-derived fungus Penicillium sp. MCCC 3A00126, along with 34 known compounds (3-36). The structures of the new compounds were determined using spectroscopic data. The absolute configuration of compound 1 was confirmed by comparing experimental and calculated ECD spectra. Cytotoxicity and ferroptosis inhibitory activities of all isolated compounds were evaluated. Compounds 14 and 15 showed potent cytotoxicity against CCRF-CEM cells, with IC50 values of 5.5 and 3.5 μM, respectively. Compounds 26, 28, 33, and 34 exhibited significant inhibitory effects on RSL3-induced ferroptosis, with EC50 values of 11.6, 7.2, 11.8, and 2.2 μM, respectively.

Summary: In this study, a series of trifluoromethyl pyrimidine derivatives were designed and synthesized. Bioassay results showed that some of these compounds exhibited higher antiviral and antifungal activities than commercial agents. In particular, compounds 5j and 5m demonstrated lower EC50 values compared to ningnan-mycin. Microscale thermophoresis experiment confirmed a strong interaction between compound 5m and TMV-CP. Moreover, compound 5u displayed significant in vitro antifungal activity against Rhizoctonia solani (RS) and effectively controlled RS in vivo.
