Summary: This study found that anhedonia severity and depression symptom severity play a central role in the network. The results suggest that anhedonia severity may predict the severity of symptom clusters in MDD, AS, ADHD, and ASD. Additionally, the severity of MDD symptoms may predict the severity of AS and ADHD symptoms. These findings indicate that anhedonia may serve as a common underlying psychopathology feature, predictive of symptom severity in depression, AS, ADHD, and ASD. Therefore, clinicians should be vigilant in screening for anhedonia and depression severity in individuals with anxiety disorders, ADHD, and/or ASD.

Summary: This study found close associations between anhedonia and suicidal ideation (SI) among adolescent patients with recurrent depressive disorder (depression). Network analysis revealed that anhedonia was directly related to SI, as well as guilt, sad mood, and motor disturbances. Sad mood and fatigue were the main bridge nodes linking anhedonia and SI. This study is important for guiding interventions to reduce the risk of SI among clinically stable adolescents with depression during the COVID-19 pandemic.

Summary: Currently, only a limited number of interventions can rapidly relieve depressive symptoms in patients with major depressive disorder or bipolar disorder. Ketamine and its enantiomer esketamine have been shown to rapidly reduce depressive symptoms in depressed patients with current suicidal ideation. Recently, esketamine has been approved for use in patients with depression at risk of suicide and for psychiatric emergency, while ketamine is increasingly used off-label. However, there is limited guidance on when and how to use these drugs in patients with depression experiencing a crisis. In this review article, the cellular and molecular mechanisms of action of ketamine and esketamine, as well as the functional brain changes following their administration are summarized. The major clinical studies on ketamine and esketamine efficacy in patients experiencing a crisis are also summarized, and a profile of patients who can benefit most from these drugs is proposed based on neurobiological and clinical observations. Additionally, the administration mode, efficacy and tolerability profiles, side effect management, possible concomitant treatments, and the issue of deprescribing are discussed.

Summary: This study examined the impact of contact with COVID-19 patients on the mental health of healthcare workers in the United States. The findings revealed that healthcare workers who provided direct care to COVID-19 patients experienced increased depressive and posttraumatic symptoms compared to those who had no contact. Higher levels of distress were also associated with more frequent contact. More comprehensive data from diverse samples of US healthcare workers are needed to fully understand the extent of this relationship.

Summary: This study investigated the magnetoencephalographic (MEG) correlates of suicidal ideation (SI) and suicide attempt history in patients with treatment-resistant major depression (TRD), both at baseline and after subanesthetic-dose ketamine infusion. The findings showed that ketamine significantly reduced SI and depression in the sample. Following ketamine infusion, attempters showed improved accuracy on the task, while non-attempters showed reduced accuracy. SI was positively associated with gamma power in frontal and parietal cortices. In an extended amygdalahippocampal region, attempters differed significantly in their emotional reactivity to angry versus happy faces as indexed by theta power differences, regardless of drug. Ketamine also reduced the association between alpha power and SI for angry compared to happy faces in a fronto-insular/anterior cingulate region important for regulating sensory attentiveness.

Summary: Rapid-acting treatments for suicidal thoughts are in high demand and have the potential to not only benefit patient populations but also provide valuable insights for suicide research methodologies. The quick onset of new interventions allows for an experimental therapeutics approach embedded with psychological and neurobiological markers to better understand the underlying pathophysiology of suicidal thoughts. Lessons learned from research on ketamine may serve as a blueprint for researchers evaluating rapid-acting treatments for suicidal thoughts, whether pharmacologic or psychotherapeutic.
