Cationic liposomes (CLs) have been widely examined as vaccine delivery nanoparticles since they can form complexes with biomacromolecules, promote delivery of antigens and adjuvant molecules to antigen-presenting cells (APCs), and mediate cellular uptake of vaccine components. CLs are also known to trigger antigen cross-presentation -the process by which APCs internalize extracellular protein antigens, degrade them into minimal CD8+ T-cell epitopes, and present them in the context of major histocompatibility complex-I (MHC-I). However, the precise mechanisms behind CL-mediated induction of cross-presentation and cross-priming of CD8+ T-cells remain to be elucidated. In this study, we have developed two distinct CL systems and examined their impact on the lysosomal pH in dendritic cells (DCs), antigen degradation, and presentation of peptide: MHC-I complexes to antigen-specific CD8+ T-cells. To achieve this, we have used 3 beta-[N-(N', N'-dimethylaminoethane)-carbamoyl] cholesterol (DC-Chol) and 1,2- dioleoyl-3- trimethylammonium-propane (DOTAP) as the prototypical components of CLs with tertiary amine groups and compared the effect of CLs and anionic liposomes on lysosomal pH, antigen degradation, and cross- presentation by DCs. Our results showed that CLs, but not anionic liposomes, elevated the lysosomal pH in DCs and reduced antigen degradation, thereby promoting cross-presentation and cross- priming of CD8+ T-cell responses. These studies shed new light on CL-mediated cross- presentation and suggest that intracellular fate of vaccine components and subsequent immunological responses can be controlled by rational design of nanomaterials.

Strengths and weaknesses of alternative noninvasive imaging approaches for microvascular ischemia

Isabella Leo, Eleni Nakou, Jessica Artico, Emmanouil Androulakis, Joyce Wong, James C. Moon, Ciro Indolfi, Chiara Bucciarelli-Ducci

Summary: Structural and functional abnormalities of coronary microvasculature are common in clinical settings and associated with worse clinical outcomes. Traditional invasive methods for assessment of coronary microvascular function have limitations and risks. Recent advances in non-invasive imaging techniques provide alternative approaches for evaluating coronary microvascular function.

Unlocking the promise of systemic STING agonist for cancer immunotherapy

Xiaoqi Sun, Xingwu Zhou, Yu Leo Lei, James J. Moon

Summary: The STING pathway is an important innate immune pathway involved in cancer immunity. Recent developments in molecules and drug delivery systems have made systemic STING agonist immunotherapy possible, showing promising tumor eradication in preclinical studies. This article discusses the potential mechanisms of STING agonism as a multifaceted anti-cancer therapy and the pharmacological challenges associated with systemic delivery of STING agonists at different levels. Drug delivery strategies to overcome these challenges are also presented and discussed. Advances in this field may unlock the potential of systemic STING agonists as effective and safe cancer immunotherapy.

Electrosprayed microparticles from inulin and poly(vinyl) alcohol for colon targeted delivery of prebiotics

Keara T. Saud, Jin Xu, Sabina Wilkanowicz, Yue He, James J. Moon, Michael J. Solomon

Summary: Recent studies have shown that inulin has a prebiotic effect by selectively promoting bacteria in the colon, modulating the gut microbiome, and generating beneficial effects on gastrointestinal inflammation, diabetes, and cancer. However, the water-soluble nature of inulin limits its prebiotic effect due to low delivery efficiency and short retention time in the colon. This study aimed to produce inulin microparticles (MPs) using the electrospray method and evaluate their material properties and bioavailability.

Accelerated DNA methylation age plays a role in the impact of cardiovascular risk factors on the human heart

Constantin-Cristian Topriceanu, Eesha Dev, Mahmood Ahmad, Rebecca Hughes, Hunain Shiwani, Matthew Webber, Kenan Direk, Andrew Wong, Martin Ugander, James C. Moon, Alun D. Hughes, Jane Maddock, Todd T. Schlegel, Gabriella Captur

Summary: This study investigates the relationship between DNAm age and A-ECG heart age, and finds that DNAm AgeAccel is associated with cardiovascular risk factors. The results suggest that participants with accelerated DNA methylation at the age of 60 may have older, weaker, and more electrically impaired hearts. Furthermore, the harmful effects of cardiovascular risk factors on cardiac age and health may be partially mediated by DNAm AgeAccel.
