Unconventional Secretion of Heat Shock Proteins in Cancer

Heat shock proteins (HSPs) are abundant cellular proteins involved with protein homeostasis. They have both constitutive and inducible isoforms, whose expression levels are further increased by stress conditions, such as temperature elevation, reduced oxygen levels, infection, inflammation and exposure to toxic substances. In these situations, HSPs exert a pivotal role in offering protection, preventing cell death and promoting cell recovery. Although the majority of HSPs functions are exerted in the cytoplasm and organelles, several lines of evidence reveal that HSPs are able to induce cell responses in the extracellular milieu. HSPs do not possess secretion signal peptides, and their secretion was subject to widespread skepticism until the demonstration of the role of unconventional secretion forms such as exosomes. Secretion of HSPs may confer immune system modulation and be a cell-to-cell mediated form of increasing stress resistance. Thus, there is a wide potential for secreted HSPs in resistance of cancer therapy and in the development new therapeutic strategies.

Summary: Melatonin has been found to have an anticancer effect on brain tumors with a negative correlation between its synthesis in the microenvironment and tumor malignancy. MT1 activation inhibits the proliferation of glioma and medulloblastoma cell lines, while MT2 activation promotes it. Tumors with a higher MT1/MT2 expression ratio show better survival rates.

Current treatment strategies include chemotherapy, radiation, aggressive surgery, and intra-peritoneal hyperthermic chemotherapy, but high recurrence rates and poor survival outcomes necessitate the urgent development of new therapeutic options. Ongoing research is focused on identifying potential therapeutic targets through comprehensive molecular profiling to improve understanding of DSRCT biology and explore novel treatment strategies in preclinical and early phase clinical trials.

Summary: Bone sarcoma is a rare disease with a low incidence in adult population. The multidisciplinary management and expert discussion of diagnostic and therapeutic strategies are essential for these patients. The Sarcoma European Latin-American Network (SELNET) aims to improve clinical outcomes in sarcoma care, with a specific focus on Latin-American countries. The Clinical Practice Guidelines (CPG) developed by SELNET provide a standardized approach to diagnose, treat, and follow-up bone sarcoma patients in the Latin-American context.

Summary: This study found that CD28-based anti-CAIX CAR PBMCs cells, releasing anti-PD-L1 IgG, showed superior antitumor effects in vivo, especially under the influence of PD-L1 via ICB. The exhaustion status of CD4 T cells, in addition to CD8 T cells, was critical for the efficacy of CAR T cells. These findings offer exciting new prospects for the treatment of refractory ccRCC and hypoxic tumors using anti-CAIX CAR T cells.

Summary: Obesity decreases life expectancy and increases age-related dysfunctions, including beta-cell dysregulation. Diluted plasma from obese human donors and sera from obese rats impair beta-cell integrity and insulin secretion, while the presence of adiponectin, a protective characteristic found in lean plasma, restores beta-cell function. Low circulating adiponectin is a key damaging element for beta-cells and modulation of the adiponectin signaling pathway could be a potential therapy for preventing age-related beta-cell dysfunction.

Establishment and Comprehensive Molecular Characterization of an Immortalized Glioblastoma Cell Line from a Brazilian Patient

Summary: This study successfully established and characterized the GBM33 cell line, which belongs to the GBM class and exhibits similar characteristics and functions as GBM cells. Compared to commercial cell lines, GBM33 cells show increased growth rate and similar sensitivity to temozolamide, and they can produce primary cilia upon serum starvation. The establishment of this cell line provides valuable tools for studying GBM biology, identifying therapeutic targets, and evaluating treatment efficacy.

Summary: This study aimed to establish a renal cell carcinoma (RCC) patient-derived xenograft (PDX) platform to identify novel clinical and molecular biomarkers of recurrence risk. Orthotopic implantation was found to be more suitable for establishing the PDX model, preserving RCC identity and major molecular alterations. The capacity of tumor engraftment was also found to be a strong prognostic indicator for patients with locally advanced disease.
