Assessment of the antioxidant activity of an olive oil total polyphenolic fraction and hydroxytyrosol from a Greek Olea europea variety in endothelial cells and myoblasts

Olive oil (OO) constitutes the basis of the Mediterranean diet, and it seems that its biophenols, such as hydroxytyrosol (HT) may scavenge free radicals, attracting distinct attention due to their beneficial effects in many pathological conditions, such as cancer. To the best of our knowedge, this is the first study in which the functional properties of an OO total polyphenolic fraction (TPF) and pure HT were examined in order to determine their antioxidant effects at a cellular level in endothelial cells and myoblasts. The test compounds were isolated using a green gradient-elution centrifugal partition chromatography-based method that allows the isolation of large volumes of OO in a continuous extraction procedure and with extremely low solvent consumption. For the isolation of HT, a combination of two chromatographic techniques was used, which is effective for the recovery of pure compounds from complex natural extracts. Moreover, TPF and HT exhibited potent free radical scavenging activity in vitro. The cells were treated with non-cytotoxic concentrations and their redox status [in terms of glutathione (GSH) and reactive oxygen species (ROS) levels] was assessed. TPF extract was less cytotoxic than HT, and the observed differences between the two cell lines used suggest a tissue-specific activity. Finally, flow cytometric analysis revealed that both TPF and HT improved the redox status by increasing the levels of GSH, one of the most important antioxidant molecules, in both endothelial cells and myoblasts, while the ROS levels were not significantly affected.

Modifiable contributing factors to COVID-19: A comprehensive review

Summary: The severe complications of COVID-19 are a result of the individual's dysfunctional immune response to the initial SARS-CoV-2 infection. Multiple toxic stressors and behaviors contribute to immune system dysfunction which the virus exploits to cause COVID-19. This study identifies eighty modifiable contributing factors that link directly to COVID-19, and shows how each factor contributes to immune system dysfunction, inflammation, coagulation, neural damage, and neurodegeneration. It emphasizes the importance of addressing and eliminating these factors in combatting COVID-19 and similar diseases caused by viral variants.

Summary: This review summarizes the existing data on the effects of toxic metals on HIF-1 signaling and its underlying mechanisms, with a special focus on the prooxidant effect of metals. The effect of metals on HIF-1 pathway varies depending on the cell type, ranging from down-regulation to up-regulation. Inhibition of HIF-1 signaling may contribute to hypoxic damage in cells, while metal-induced activation may promote tumor growth and contribute to the carcinogenic effects of heavy metals.

Summary: The current risk assessment approach for chemicals fails to consider real-life exposure scenarios. The exposure to chemical mixtures in everyday life has become a concern in recent years. This study investigated the long-term effects of exposure to a mixture of 13 chemicals in adult rats and found dose-dependent changes in all examined organs. The main organs involved in biotransformation and clearance consistently presented histopathological changes. Exposure to very low doses of the tested mixture for 18 months induced histopathological lesions and cytotoxic effects in a dose and tissue-dependent manner.

Species composition and ecological structure of ground beetles (Coleoptera, Carabidae) communities as biological indicators of the agro-environmental sustainability

Summary: The detrimental environmental consequences caused by intensification of crop cultivation can be prevented through monitoring of specific biological indicators. This study focused on the impact of crop type and cultivation intensity on the community of ground beetles in the forest-steppe of Western Siberia. The ground beetles' community showed a high evenness of species distribution across the agroecosystems. Differences in ground beetle distribution were observed between wheat and corn crops. The use of bioindicators allowed for assessing the environmental sustainability of the agricultural environment and provided insights for ecologically directed correction of agrotechnological operations in agroecosystem management.

Summary: The aim of our study was to evaluate the differences among different groups of drug users, focusing on hematological, biochemical, urinary, and cardiological aspects. A retrospective study was conducted on 2572 hospitalized patients in Bucharest, Romania from 2015 to 2021. The study group was characterized based on demographic data, personal history/co-morbidities, consumption history, and substitution treatment. The analysis of medical records revealed correlations between GGT levels and alcohol intake as well as contagious diseases, a decrease in MCH and MCHC levels across the entire study group, and correlations between specific cardiological events and certain substance groups. The study provides important information on different types of drug users and supports future research on the chemical structures of consumed compounds.

Psoriasis immunometabolism: progress on metabolic biomarkers and targeted therapy

Summary: Psoriasis is a common inflammatory disease that mainly affects the skin and can be associated with comorbidities such as psoriatic arthritis, Crohn's disease, metabolic syndrome, and cardiovascular disease. The development of psoriasis involves complex interactions between keratinocytes, T helper cells, and immune cells in the skin. Understanding the metabolic reprogramming of these cells in psoriatic skin can provide insights into novel diagnostic and therapeutic targets. The dysregulation of metabolic pathways, such as glycolysis, TCA cycle, amino acid metabolism, and fatty acid metabolism, plays a significant role in psoriasis. Targeting these pathways and restoring metabolic imbalances may present promising therapeutic opportunities for long-term management of psoriasis.

Summary: Human milk is the healthiest option for newborns, but infant formula is a valuable alternative in certain circumstances. Ensuring pollutant-free composition, infant formulas are regulated and monitored for safe exposure. While legislation varies across countries, standard policies for protecting vulnerable infants are in place. This article presents current regulations and directives for limiting endocrine-disrupting chemicals and persistent organic pollutants in infant formulas, highlighting the need for limited risk assessment studies to assess the health risks for infants from dietary exposure to pollutants.

Summary: This study aimed to investigate the impact of carnosic acid (CA) on indomethacin (IND)-induced gastric ulceration. The results showed that CA pretreatment attenuated both gross morphological lesions and histopathological alterations induced by IND, and it achieved gastroprotective effects by reducing oxidative stress and suppressing inflammation.

Summary: The wide-scale emergence of glyphosate-resistant weeds has caused an increase in the application of herbicide mixtures containing glyphosate plus dicamba or glyphosate plus 2,4-D. This raises concerns about the environmental and health risks from exposure to a mixture of herbicide active ingredients. The study evaluated the hepatotoxic effects of perinatal exposure to glyphosate alone or in combination with 2,4-D and dicamba on Wistar rats. The findings highlight the importance of assessing the combined effects of chemical pollutants at safe individual doses.
