Summary: Endothelial cells play crucial roles in blood transport, vessel homeostasis, inflammatory responses, transendothelial migration, and blood vessel formation. CD34 is commonly used as an endothelial marker, but its expression and function vary in different organs and pathological conditions. This study investigates the expression and function of CD34 in endothelial cells, revealing that CD34(high) cells have lower proliferation but higher expression of IL-33 and Angiopoietin 2 compared to CD34(low) cells. CD34(high) endothelial cells also enhance the proliferation of regulatory T lymphocytes, suggesting their immunogenic ability and potential role in tolerogenic response.

Summary: The physical properties of nanoparticle-based vaccines, such as their shape, play a crucial role in antigen capture, lymphatic delivery, antigen presentation, and immune response in lymph nodes. Star-shaped gold nanoparticles (AuNPs) were found to capture and retain more repetitive antigen epitopes. Rods and star-shaped nanovaccines primarily drained into the lymph node follicles region, while cage-shaped nanovaccines showed stronger paracortex retention. Surprisingly, star-shaped nanovaccines elicited potent humoral immunity, preventing tumor growth in prophylactic studies. Cage-shaped nanovaccines preferentially presented peptide-MHC I complexes and showed the strongest therapeutic efficacy when combined with PD-1 checkpoint inhibitors in established tumor studies.

Summary: The study revealed specific T cell responses against CTAs and TAAs in HCC patients. In early-stage HCC patients, SALL4-specific response was the most prominent, followed by MAGE-A3, NY-ESO-1, MAGE-A1, and SSX2. The protective role of CTA-specific responses was suggested for one-year recurrence-free survival after treatment. As HCC progressed, CTA and SALL4-specific T cell responses decreased while AFP-specific response increased, indicating the potential of immunotherapy at an early stage of HCC.
