Summary: This study compared the risk discrimination for predicting CHD and CVD deaths between the Pooled Cohort Equations (PCE), the MESA Risk Score (with and without CAC), and the addition of CAC to the PCE. The results showed that adding CAC to the PCE improved risk discrimination, and the MESA Risk Score with CAC, as well as the PCE plus CAC, performed best among patients with 5% to 20% estimated risk. Additionally, CAC also modestly improved discrimination in low- and high-risk groups.

Summary: The aim of this study was to investigate the association of a genetic depression risk score (GDRS) with mortality (all-cause and cardiovascular) and markers of depression in patients referred for coronary angiography. The results showed that GDRS was significantly associated with all-cause and cardiovascular mortality. However, it was not associated with antidepressant use or a history of depression.

Summary: The aim of this study was to investigate the use of the European SCORE model in a dental setting and its association with periodontitis. It was found that both periodontitis patients and non-periodontitis controls had a significant frequency of 'high' and 'very high' 10-year CVD mortality risk. The presence and extent of periodontitis, along with other factors, were found to be significant risk indicators for a 'very high' SCORE category.

Summary: This study aimed to investigate the predictive ability of polygenic risk scores (PRSs) for cardiovascular (CV) mortality, and the additive effect between PRS and lifestyle behavior. The findings suggest that PRS for coronary artery disease (CAD) and type 2 diabetes (T2DM) are significantly associated with CV mortality, and adherence to an unfavorable lifestyle further increases the risk. Furthermore, 32.1% of CV mortality is attributable to lifestyle behavior, highlighting the importance of lifestyle modification in preventing CV mortality.

Summary: Cardiovascular disease is the leading cause of death globally, with coronary artery calcium score being a powerful method in its assessment. Romania has a high risk of cardiovascular diseases but limited data on calcium score prevalence. A retrospective study found that age, dyslipidemia, hypertension, smoking, and diabetes were associated with different levels of coronary artery calcification, with smoking and diabetes being the strongest predictors for very severe calcification. Patients with multiple risk factors were significantly more likely to have very severe calcification.

Summary: Multi-omic profiling of patients with coronary artery disease reveals links between alterations in the serum metabolome with genetics, diet, and the microbiome. The study found that these metabolomic alterations are associated with metabolic impairment preceding clinically overt coronary artery disease. The results highlight the importance of understanding risk-factor heterogeneity in coronary artery disease through the serum metabolome.

Summary: The GENEMACOR study investigated the main environmental and genetic risk factors for coronary artery disease (CAD) and evaluated the role of a genetic risk score (GRS) in disease prediction, discrimination, and reclassification. The results showed that both traditional risk factors and genetic variants influenced the likelihood of CAD, and incorporating GRS into the traditional model improved the predictive capacity, discrimination, and reclassification of the disease. These findings have important implications for clinical diagnosis and treatment.
