Summary: Pretreatment of Dendrobium officinale with organic solvents may enhance the immunostimulatory activity of polysaccharides and affect the mannose/glucose ratio, with DOOS-CP showing stronger activity than DO-CP. Polysaccharides from puffed Dendrobium officinale exhibited significantly lower activity, with the most active polysaccharide containing over 95% neutral sugar and a mannose/glucose ratio of 3.0. Additionally, DOOS-CP was a refined fraction with a major peak representing a molecular weight of 250 kDa.

Summary: cDFP-W1 is a new polysaccharide isolated from the stems of Dendrobium fimbriatum Hook, with high immunostimulatory activities. In vivo and in vitro experiments showed that cDFP-W1 can significantly enhance the immune response in mice by increasing the proportions of various immune cell subpopulations and promoting cell proliferation and phagocytosis. The immunostimulatory effects of cDFP-W1 are closely associated with the activation of MAPKs, NF-kappa B, and PI3K/Akt signaling pathways, as indicated by western blot analysis.

Summary: Pegylated capsule depolymerase (PEG-CapD-CPS334C) showed improved pharmacokinetic and therapeutic properties in treating mice infected with Bacillus anthracis. The treatment protected mice from infection and did not rely on antibiotics. These findings suggest that enzyme-catalyzed removal of the capsule may be a potential therapeutic strategy for multidrug-resistant anthrax and other bacterial infections.

Summary: Photodynamic therapy (PDT) eradicates tumors by generating toxic reactive oxygen species (ROS) using a photosensitizer (PS) and appropriate light. We enhanced the immune response to tumors after PDT by using a biocompatible herb polysaccharide as the carrier of the PS. The use of Dendrobium officinale polysaccharide (DOP) and positively charged nanoparticles significantly improved the immune response after PDT.

Summary: In this study, an oligosaccharide sequencing analysis was used to successfully correct the backbone of Dendrobium officinale polysaccharide (DOP) to mannan. This analysis involved producing oligosaccharide fragments through mild acid hydrolysis of DOP followed by p-aminobenzoic ethyl ester (ABEE) derivatization. The isolated and identified oligosaccharides with varied polymerization degrees (DP = 2-10) confirmed that the backbone of DOP is a beta-D-1,4-mannan. This oligosaccharide analysis is more straightforward and reliable compared to conventional methods.

Summary: Crosslinking affects the mechanical behavior and tissue metabolism of collagen-based tissues. Aging and diabetes alter the crosslinking type and density, thus changing tissue properties. High-connectivity enzymatic crosslinking and advanced-glycation end-products result in uniform deformation under daily activity, while low-connectivity enzymatic crosslinking does not. High-connectivity model displays more sliding, while AGEs induce instability in structures near the binding sites.
