High performance and toxicity assessment of Ta3N5 nanotubes for photoelectrochemical water splitting

Kaiqi Xu, Athanasios Chatzitakis, Sanne Risbakk, Mingyi Yang, Paul Hoff Backe, Mathieu Grandcolas, Magnar Bjoras, Truls Norby

Summary: In this study, Co-based cocatalysts were electrodeposited on mesoporous Ta3N5 nanotubes to achieve a photocurrent density of 6.3 mA/cm(2) at 1.23 V vs. SHE under simulated solar illumination. The best performing electrode not only exhibited high photocurrent density, but also improved stability under intense photoelectrochemical water splitting conditions. The dual function of the cocatalyst in enhancing both photoelectrochemical performance and stability was highlighted.

Pairwise relatedness testing in the context of inbreeding: expectation and variance of the likelihood ratio

Hilde Kjelgaard Brustad, Magnus Dehli Vigeland, Thore Egeland

Summary: This paper investigates the effects of inbreeding on the likelihood ratio (LR) in forensic kinship testing, extending previous formulas to cover all possible pairwise relationships by using Jacquard coefficients. It also highlights the often overlooked bias from background inbreeding in forensic kinship computations, showing how it can be modeled as a continuous variable and impact LR results significantly in specific cases.

Making decisions in missing person identification cases with low statistical power

Franco L. Marsico, Magnus D. Vigeland, Thore Egeland, Mariana Herrera Pinero

Summary: The study proposes a general strategy for dealing with missing person identification cases through DNA-database search, focusing on the identification of abducted children in Argentina during a civic-dictatorship. The researchers provide a statistical method for selecting likelihood ratio thresholds and offer an open-source software for computing LR thresholds and error rates. This strategy could be applied to other large-scale DNA-based identification cases with low statistical power.

Summary: Sturge-Weber syndrome (SWS) is a neurocutaneous disorder characterized by vascular malformations, often caused by a somatic missense mutation in the GNAQ gene. This study identified a novel somatic mutation in the GNB2 gene in one SWS patient, which affects G-protein complex reassembly and downstream signaling pathways. Both mutations decreased cellular proliferation and YAP synthesis, suggesting a key role for the Hippo signaling pathway in SWS vascular pathogenesis.

Joint DNA-based disaster victim identification

Magnus D. Vigeland, Thore Egeland

Summary: This study focuses on computational and statistical aspects of DNA-based victim identification after disasters. By jointly identifying all victims using complete genetic data, the issues of suboptimal individual identification and inconsistencies in statistical summaries are addressed. The method of deriving individual identification probabilities from joint solutions improves efficiency and handles realistic cases effectively.

abc4pwm: affinity based clustering for position weight matrices in applications of DNA sequence analysis

Omer Ali, Amna Farooq, Mingyi Yang, Victor X. Jin, Magnar Bjoras, Junbai Wang

Summary: The abc4pwm tool efficiently clusters PWMs from multiple sources, generates representative motifs for each cluster, evaluates clustering quality automatically, and filters out incorrectly clustered PWMs. It can also automatically update the human DNA-Binding Domain (DBD) family database, classify known human TF PWMs to the respective DBD family, and conduct TF motif searching and motif discovery using a new ensemble learning approach.

A Novel Nonsense Variant in GRM1 Causes Autosomal Recessive Spinocerebellar Ataxia 13 in a Consanguineous Pakistani Family

Hammad Yousaf, Ambrin Fatima, Zafar Ali, Shahid M. Baig, Mathias Toft, Zafar Iqbal

Summary: This study reports a novel pathogenic variant in the GRM1 gene in a consanguineous Pakistani family with partially overlapping clinical features of SCAR13. In addition, the family presented quadrupedal gait and non-progressive symptoms, which have not been recognized previously.

Summary: Intellectual developmental disorder with paroxysmal dyskinesia or seizures (IDDPADS) is a rare childhood-onset movement disorder characterized by paroxysmal dyskinesia, developmental delay, impaired cognition, progressive psychomotor deterioration, and drug-refractory seizures. In this study, we identified a novel missense variant in the Phosphodiesterase 2A (PDE2A) gene in three consanguineous Pakistani families with overlapping phenotypes. Haplotype analysis revealed a shared haplotype among the families, suggesting a founder effect in this region. Our findings expand the clinical and mutation spectrum of PDE2A-related recessive disease.

Summary: The objective of this study is to identify the genetic causes of rare spinocerebellar disorders in the Pakistani population. Whole exome sequencing was used to investigate nine consanguineous families, resulting in the identification of six novel pathogenic variants and three already reported pathogenic variants. This study expands the mutation spectrum of rare spinocerebellar disorders and highlights the usefulness of next-generation sequencing in combination with clinical investigation for better diagnosis.

Summary: Long-term use of antibiotics has a lasting impact on gene expression and DNA methylation, leading to potential side effects. This study found that patients receiving amoxicillin treatment for 100 days showed changes in gene expression and DNA methylation, while placebo group showed minimal changes. After 100 days, 28 genes, including 19 immunoglobulin genes, were significantly differentially expressed. These expression levels were still not completely restored at one-year follow-up. Significant changes in DNA methylation mainly occurred between 100 days and one-year follow-up. Hence, long-term amoxicillin treatment can result in lasting alterations in both gene expression and DNA methylation.

Summary: Novel antimicrobial polymer CS/PPC NPs were synthesized by reacting mono-/bis-epoxy capped PPC with chitosan, showing potential antibacterial activity against both Gram-positive and Gram-negative bacteria. The chemical structure, particle size, and surface charge were characterized using various techniques. The minimum inhibitory concentration strongly depended on the nature of the epoxy-imine network and the feed ratio of functional groups between PPC and chitosan.
