Molecular cloning, biological effect, and tissue distribution of interleukin-8 protein in mandarin fish (Siniperca chuasti) upon Flavobacterium columnare infection

Interleukin-8 (IL-8), a CXC-type chemokine, plays a key role in acute inflammation by recruiting neutrophils in mammals. In the present study, the open reading frame (ORF) of IL-8, encoding 99 amino acids was cloned in mandarin fish, and its function in inflammation was investigated. The IL-8 contains four conserved cysteine residues, with the first two forming the CXC signature motif. The genomic sequence of mandarin fish IL-8 has four exons and three introns, a typical gene organization of the CXC chemokine. Bioactive recombinant IL-8 (rIL-8) exhibited a,chemotactic effect on head kidney leukocytes in vitro, and activates the transcription of the inflammatory genes, IL-8 and IL-10. When mandarin fish was challenged intraperitoneally with the pathogenic bacterium Flavobacterium columnare G(4), the steady-state protein level of IL-8 was up-regulated in trunk kidney and head kidney. These results suggest that IL-8 is a functional CXC chemokine in mandarin fish, and plays a key role in the inflammatory responses towards bacterial infection. (C) 2017 Elsevier Ltd. Crosstalks between NOD1 and Histone H2A Contribute to Host Defense against Streptococcus agalactiae Infection in Zebrafish

Summary: In this study, the functional correlation between NOD1 and histone H2A variant in response to Streptococcus agalactiae infection was reported. The deficiency of NOD1 promoted S. agalactiae proliferation and decreased larval survival in zebrafish. The results highlight the importance of NOD1 deficiency in immune-related and metabolic pathways regulation, and the correlation between zebrafish NOD1 and histone H2A variant in defense against S. agalactiae infection.

Structure, function and immune evasion strategies of aquareoviruses, with focus on grass carp reovirus

Summary: AqRVs are a type of virus that can infect various wild or cultured bony fishes, causing serious muscle and viscera haemorrhage syndrome and significant economic losses worldwide. Research on the viral proteins of AqRVs has revealed their roles in RNA transcription, virion assembly, cell attachment, and viral replication. Additionally, the structural and functional studies of AqRVs have provided insights into their replication and immune evasion strategies, leading to the development of vaccines and antiviral agents for GCRV, the most pathogenic AqRV.

The negative regulation of Vdrs during viral infection can be prevented by vitamin D treatment, enhancing resistance against GCRV.
