Citrus nomilin down-regulates TNF-α-induced proliferation of aortic smooth muscle cells via apoptosis and inhibition of IκB

Nomilin is a bitter compound present in citrus and has been demonstrated as useful for various disease preventions through anti-proliferative, anti-inflammatory, and pro-apoptotic activities. Although in vitro disease models have shown that certain limonoids in the p38 mitogen-activated protein kinase signal cascade, the downstream signaling pathways remain unclear. In this study, the effects of nomilin on the proliferation and apoptotic pathways of human aortic smooth muscle cells (HASMCs) that forms the basis of progression of atherosclerotic diseases and restenosis was tested for the first time. The cellular uptake level and stability of nomilin were determined by high-performance liquid chromatography and high-resolution mass spectra. Pretreatment of HASMCs with nomilin stimulated extrinsic caspase-8, intrinsic caspase-9, and apoptotic caspase-3 and resulted in significant inhibition of TNF-alpha-induced proliferation. Additionally, results showed a decreased ratio of anti-apoptotic Bcl-2 protein to pro-apoptotic Bax (Bcl2/Bax), indicating mitochondrial dysfunction consistent with apoptosis. Furthermore, nomilin significantly decreased the phosphorylation of I kappa B alpha, an inhibitor of NF-kappa B and subsequently, reduced the downstream inflammatory signaling in TNF-alpha treated HASMCs. Our findings indicate that the anti-proliferative activity of nomilin on TNF-alpha-induced HASMCs results from apoptosis through a mitochondrial-dependent pathway and suppression of inflammatory signaling mediated through NF-kappa B.

Recent Advances in Intrahepatic Biliary Epithelial Heterogeneity

Summary: Biliary epithelium, also known as cholangiocytes, is a heterogeneous population of epithelial cells in the liver with different responses and functions depending on their size and location in the biliary tract. Recent findings have shown that the intrahepatic biliary tree exhibits heterogeneity in terms of morphology, function, hormone expression and signaling, response to injury, and roles in liver regeneration. This review summarizes the significant characteristics and differences between small and large cholangiocytes, as well as the in vitro and in vivo models used to evaluate heterogeneity. Future studies elucidating the role of biliary heterogeneity in liver diseases characterized by ductular reaction may provide new therapeutic approaches.

Conjugated Bile Acids Promote Lymphangiogenesis by Modulation of the Reactive Oxygen Species-p90RSK-Vascular Endothelial Growth Factor Receptor 3 Pathway

Summary: Conjugated bile acids (BA) are elevated in liver pathologies and metastatic lymph nodes. This study investigates the effects of BAs on lymphangiogenesis. The results indicate that BAs promote lymphatic network expansion and enhance LEC invasion, migration, and tube formation through ROS-mediated activation of p90RSK and upregulation of VEGFR3. Furthermore, BAs induce YAP expression, a regulator of lymphangiogenesis. Overall, this study establishes a novel mechanism by which BAs promote lymphangiogenesis.

Oxidative Stress-Induced Liver Damage and Remodeling of the Liver Vasculature

Summary: The liver plays a critical role in targeting and clearing foreign material, but stress-induced factors can disrupt its normal functioning and lead to liver diseases such as steatosis, inflammation, fibrosis, cirrhosis, and liver cancers. Oxidative stress and inflammatory signaling play a key role in this process, causing mitochondrial dysfunction, increased reactive oxygen species expression, and depletion of ATP. Changes in the liver vasculature also contribute to the pathogenesis of these diseases.

Accelerated solvent extraction of red onion peel extract and its antimicrobial, antibiofilm, and quorum-sensing inhibition activities against Listeria monocytogenes and Chromobacterium violaceum

Summary: In this study, we extracted flavonoids and anthocyanins from dried red onion peels using accelerated solvent extraction (ASE) to produce red onion peel extract (ROPE). High-performance liquid chromatography analysis identified important flavonols and anthocyanins in ROPE. The total flavonoid content of ROPE was 459.15 mg/g +/- 16.45 (DW), while the total anthocyanin content was 33.14 mg/g +/- 0.99 (DW). ROPE showed strain-specific effects against L. monocytogenes, inhibiting biofilm formation, motility, and quorum sensing in a concentration-dependent manner. The results suggest that red onion peel waste has the potential to be a source of natural antibacterial compounds for food safety in the circular economy.
