Summary: Recent studies suggest that X-linked glucose-6-phosphate dehydrogenase (G6PD) deficiency is associated with a proinflammatory state. This study aimed to investigate the enzyme activity and inflammatory markers in relation to G6PD deficiency and X inactivation. The results showed that G6PD deficiency may be associated with increased inflammatory markers, especially in elderly females, and worsened by asymmetric X-chromosome inactivation.

Summary: Replenishing nucleotide pools is essential for supporting cell proliferation in cancer cells. Glutamine and the oxidative branch of the pentose phosphate pathway (ox-PPP) are key in nucleotide synthesis and maintaining redox status. Inhibiting G6PD and depriving glutamine can decrease colon cancer cell proliferation and induce cell cycle arrest and apoptosis. This study highlights the potential of targeting oxidative PPP enzymes and glutamine catabolism in combating colon cancer.

Summary: This study compared the risk of intracranial atherosclerotic stenosis (ICAS) in stroke patients with and without glucose-6-phosphate dehydrogenase (G6PD) deficiency in a Chinese cohort. The results showed that stroke patients with G6PD deficiency had a higher prevalence and burden of ICAS, particularly in male, older, and hypertensive patients.

Summary: This article reports a patient with chronic hemolysis who has a mutation at position 219 in the G6PD protein. Structural and biochemical experiments show that this mutation significantly reduces catalytic activity, stability, and dimer formation of the mutant enzyme. These findings have implications in the development of small molecule activators for treating related mutations.

Summary: Vitamin E has antioxidant effects on erythrocytes, protecting glutathione from oxidation. This systematic review found that Vitamin E significantly improves hemoglobin levels and decreases reticulocyte levels in patients with chronic and acute hemolysis. Furthermore, it improves packed cell volume, red blood cell half-life, and decreases reticulocyte levels in before and after studies.

Summary: Glucose-6-phosphate dehydrogenase (G6PD) is a key enzyme in the pentose phosphate pathway, responsible for producing a reducing agent necessary for maintaining reduced glutathione levels. This study investigated the effects of certain drugs on G6PD enzyme activity using purification and in vitro/in silico methods. The results showed that dobutamine hydrochloride significantly reduced enzyme activity by causing structural alterations in substrate and coenzyme binding sites.
