In this work, high surface area mesoporous silica (SBA-15) was loaded with paclitaxel (taxol, PTX) and was further entrapped into poly(lactic acid-co-glycolic acid) (PLGA) microparticles (MPs). A modified solvent evaporation-emulsion method was used in order to formulate the composite microparticles with sizes of 8-12 mu m. PTX loaded SBA-15 as well as the PLGA/PTX-SBA-15 composites were characterized in terms of their morphology, crystal structure and thermal properties. Drug content, loading efficiency, particle size and the in-vitro drug release kinetics of the PLGA/PTX-SBA-15 microspheres were also investigated. The in vitro release studies Were carried out using Simulated Body Fluid (SBF) at 37 degrees C revealing that the prepared formulations present higher dissolution rate than pure PTX and sustained pattern which is ideal for anticancer carriers. Modeling and data analysis of the in vitro drug release was also investigated. It was also shown that all microparticles have low cytotoxicity in HUVE cells. Finally, it was found that drug loaded microparticles are very effective in Human Cervical Adenocarcinoma (HeLa) cells. (C) 2016 Elsevier B.V. All rights reserved.

AOP-Based Transformation of Abacavir in Different Environments: Evolution Profile of Descyclopropyl-Abacavir and In Silico Toxicity Assessment of the Main Transformation Products

Eleni Evgenidou, Konstantina Vasilopoulou, Lelouda-Athanasia Koronaiou, George Kyzas, Dimitrios Bikiaris, Dimitra Lambropoulou

Summary: This study investigates the photocatalytic transformation of the antiviral drug abacavir using different advanced oxidation processes (AOPs) and assesses their efficiency in removing abacavir in different matrices. The results show that all processes are effective in eliminating abacavir within a short time, but the presence of organic matter or inorganic constituents can hinder the process. The study also identifies major transformation products and their evolution profiles in different matrices, indicating different transformation pathways dominate in each matrix. The toxicity prediction of the major transformation products suggests that only hydroxylation can detoxify the treated solution.

Giulia Fredi, Edoardo Zonta, Alessandro Dussin, Dimitrios N. Bikiaris, George Z. Papageorgiou, Luca Fambri, Andrea Dorigato

Summary: This study successfully prepared and characterized polylactide/poly(propylene 2,5-furandicarboxylate) (PLA/PPF) and polylactide/poly(butylene 2,5-furandicarboxylate) (PLA/PBF) blends in bulk and fiber forms, and investigated the influence of poly(alkylene furanoate) (PAF) concentration and compatibilization on their physical, thermal, and mechanical properties. Both blends were successfully compatibilized by Joncryl, which improved the interfacial adhesion and reduced the size of PPF and PBF domains. PBF was found to effectively toughen PLA in bulk samples, while PPF showed no significant plasticizing effects. In fiber samples, both PPF and PBF exhibited plasticizing effects, resulting in significantly higher strain at break values compared to neat PLA.

The use of polymorphic state modifiers in solid lipid microparticles: The role of structural modifications on drug release performance

Serena Bertoni, Elena Simone, Stefano Sangiorgi, Beatrice Albertini, Nadia Passerini

Summary: This study investigated the correlation between the structure and release properties of solid lipid microparticles (MPs) with different liquid additives. The additives accelerated the conversion of the unstable alpha-form of tristearin to the stable beta-polymorph and caused structural modifications in the MPs. The presence of additives prolonged the drug release in water and resulted in higher release profiles in biorelevant media. The findings suggest that the release behavior can be influenced by the polymorphism and supramolecular-level structural modification of lipid formulations containing crystal modifiers.
