Aging increases the susceptibility of cisplatin-induced nephrotoxicity

Cisplatin (CDDP) nephrotoxicity is one of the most common side effects in cancer treatment, causing the disruption of chemotherapy. In this study, we analyzed the influence of nongenetic factors on CDDP-induced nephrotoxiciy using the data from 182 CDDP-treated and 52 carboplatin (CBP)-treated patients. The mean change of eGFR (100 % to baseline) in CDDP-treated patients was -9.2 %, which was significantly lower than that in the population with CBP therapy. By using the chi-squared test and multivariate logistic regression analysis, age (>= 50 years) is found associated with CDDP-induced nephrotoxicity, with odds ratio (OR) of 9.167 and 11.771, respectively. Three-and 18-month-old mice were employed to study the age-dependent susceptibility of CDDP-induced nephrotoxicity. Biochemical parameters, histopathogical examination, and mRNA biomarkers indicated that old mice were subjected to more severe kidney injury. In addition, old mice accumulated more CDDP in kidney than young mice, and the protein level of CDDP efflux transporter, MATE1, in aged mice kidney was 35% of that in young mice. Moreover, inflammatory receptor TLR4 was higher in the kidney of old mice, indicating the alteration of inflammatory signaling in old mice. After CDDP administration, the induced alterations of TNF-alpha, ICAM-1, and TLR4 were more extensive in old mice. To summarize, aging increased the susceptibility of CDDP-induced renal function decline or nephrotoxicity.

Summary: This study found that LINC-PINT is significantly downregulated in nasopharyngeal cancer tissues compared to rhinitis tissues, and low LINC-PINT expressions are associated with poorer prognosis in patients receiving radiotherapy. LINC-PINT plays a functional role in inhibiting malignant phenotypes and sensitizing cancer cells to irradiation in vitro and in vivo. Mechanistically, LINC-PINT inhibits DNA damage repair through the ATM/ATR-Chk1/Chk2 signaling pathways and increases radiosensitivity by interacting with DNA-PKcs.

The protective effect of rabeprazole on cisplatin-induced apoptosis and necroptosis of renal proximal tubular cells

Linc00312 Single Nucleotide Polymorphism as Biomarker for Chemoradiotherapy Induced Hematotoxicity in Nasopharyngeal Carcinoma Patients

Summary: This study explores the relationship between genetic variations of linc00312 and the risk of chemoradiotherapy induced toxic reactions in nasopharyngeal carcinoma (NPC) patients. The results show that linc00312 rs15734 is significantly associated with severe leukopenia, while rs12497104 is associated with thrombocytopenia. These genetic variations may serve as biomarkers for personalized medicine in NPC patients.

Structural Analysis and Prediction of Hematotoxicity Using Deep Learning Approaches

Additionally, the study utilized SHAP and atom heatmap methods to identify important features and structural fragments related to hematotoxicity, and employed MMPA and representative substructure derivation technique to further investigate the transformation principles and distinctive structural features of hematotoxic chemicals.

Epigallocatechin Gallate Attenuates Gentamicin-Induced Nephrotoxicity by Suppressing Apoptosis and Ferroptosis

Summary: This study demonstrated that epigallocatechin gallate (EG) from green tea attenuated nephrotoxicity induced by gentamicin (GEN), reducing the increase of ROS and MDA and the decrease of GSH levels, and activating the protein levels of Nrf2 and HO-1. Furthermore, EG alleviated apoptosis and ferroptosis caused by GEN. However, the protective effects of EG were weakened when Nrf2 was knocked down.

The ferroptosis signature predicts the prognosis and immune microenvironment of nasopharyngeal carcinoma

Summary: This study explored the dysregulated expression of ferroptosis-related genes (FRGs) between normal control and tumor samples in nasopharyngeal carcinoma (NPC). Four FRGs (ABCC1, GLS2, CS, and HMGCR) were identified as associated with poor prognosis in NPC patients. A risk model using two FRGs (ABCC1 and GLS2) showed good predictive performance according to the ROC curve. Multivariate analysis indicated that the risk score and intratumoral TILs were independent risk factors for prognosis.

Summary: The study elucidated the underlying mechanism of FAS-AS1 and its genetic variation rs6586163 in nasopharyngeal carcinoma (NPC). The variant genotype carriers of rs6586163 were associated with a lower risk of NPC and better overall survival. FAS-AS1 was downregulated in NPC tissues and overexpression of FAS-AS1 was associated with early clinical stage and better treatment efficacy.

Migration inhibition and selective cytotoxicity of cobalt hydroxide nanosheets on different cancer cell lines
