Summary: Dot blot assays have traditionally used antibodies as the primary molecular recognition element, but now aptamers can also be utilized. These assays are often used to monitor SELEX progress and assess the potency of aptamer candidates. Efforts to expand aptamer-based dot blot assays for diagnostic purposes are currently slow, suggesting a need for further development.

Summary: This study evaluated various parameters affecting the performance of gold nanoparticles as labels, finding that increasing particle size enhances antibody adsorption efficiency. The functionality of antibody-GNPs against morphine and the accuracy of the Morphine-BSA structure were confirmed through dot-blot assay. Lastly, the effect of gold nanoparticle size on the limit of detection in ICA was assessed.

Summary: Fungicide abuse can lead to the emergence of fungicide-resistant fungal pathogens, posing a threat to agriculture and food safety. In this study, researchers developed iARMS, an isothermal amplification refractory mutation system, which enables rapid and sensitive detection of fungicide-resistant crop fungal pathogens. The iARMS assay showed high specificity and sensitivity, and it was able to detect rare fungicide-resistant isolates.

Summary: The COVID-19 pandemic has led to increased focus on clinical assays, particularly serological testing, to assess exposure to SARS-CoV-2 and immune response to the virus and vaccines. While some tests faced quality issues, they still contribute to public health efforts, highlighting challenges for both emerging technologies and established platforms.

Summary: In this study, a Western blot strip coated with multiple SARS-CoV-2 antigens was developed to comprehensively study antibody profiles in COVID-19 patient sera. The levels of specific antibodies directed against SARS-CoV-2 spike, S2, and nucleocapsid proteins showed a positive correlation with disease progression. The multi-antigen-coated Western blotting assay also demonstrated a correlation with the neutralizing potency of the sera. Additionally, a unique antigenic epitope was identified within the S2 subunit that was specifically recognized by COVID-19 patient serum.

Summary: In this study, a rapid and user-friendly dot-blot assay for the detection of ochratoxin A was developed using peptide conjugated gold nanoparticles as a detection agent. The assay exhibited high binding affinity towards ochratoxin A and had a low limit of detection. The dot-blot assay showed comparable results with the conventional high performance liquid chromatography in the detection of ochratoxin A in wheat samples. This portable and easy-to-use assay has the potential for application at different stages of food production and distribution to ensure safe and healthy food.

Summary: The study evaluated the diagnostic accuracy of serological tests for COVID-19, finding that ELISA and CLIA tests performed better in terms of sensitivity than LFIA. IgG-based tests had higher sensitivity than IgM-based tests, with combined IgG/IgM test-based CLIA tests showing the best overall diagnostic test accuracy.

Summary: A speedy, sensitive, accurate, and user-friendly colorimetric assay was developed for the diagnosis of antibiotic resistance. The combination of the peroxidase-like property of Au-Fe3O4 nanozyme and the specific gene identification capability of CRISPR-Cas12a enables the detection of antibiotic resistance genes. This method allows for the flexible and efficient surveillance and control of microbial water quality by capturing and analyzing diagnostic signals with a smartphone.

Summary: To monitor infectious diseases, it is important to assess immunoreactivity and measure different antibody isotypes at different stages of the immune response. Lyme borreliosis can be caused by various Borrelia species, so correct classification of samples requires evaluating immunoreactivity against different antigens. This study demonstrates a dual-reporter multiplex immunoassay that allows simultaneous evaluation of IgG and IgM immunoreactivity against different bacterial antigens, providing more serological information in less time.

Summary: The ongoing coronavirus pandemic has posed significant challenges to humanity, affecting not only personal health but also the environment. This review examines the littering of face masks and personal protective equipment and their subsequent environmental impact in terms of microplastic and microfiber pollution. The findings highlight a substantial increase in the usage of personal protective equipment, particularly face masks, and the consequent mismanagement leading to pollution in the marine environment. Synthetic fibers used in disposable and reusable masks contribute to microfiber pollution, with the latter releasing a higher amount of microfibers due to repeated laundry and disinfection. Limited studies have addressed the release of microfibers from masks, and no studies have reported on the impact of personal protective equipment. These results emphasize the urgent need for critical analysis of pandemic-related littering and microfiber release, as well as the implementation of environmental management plans worldwide.

Summary: There are differences in hematocrit (HCT) changes among men on different forms of testosterone therapy (TTh). Intranasal testosterone therapy does not appear to have a significant impact on HCT compared to longer-acting forms of TTh, while intramuscular testosterone and subcutaneous testosterone pellet therapy are associated with an increase in HCT.
