In situ mass spectrometry analysis was conducted for the hydrothermal decomposition of mannitol at different temperatures and residence times in a batch reactor. The temperature was varied in the range of 140-220 degrees C, and the residence time was varied between 5 and 20 min. The products of mannitol decomposition included furfural, 5-hydroxymethylfurfural, phenol, formic acid, and levulinic acid. At high temperatures, the mass spectrum showed peaks at m/z 46 and 116 that corresponded to formic acid and levulinic acid, respectively. Conventional high-performance liquid chromatography could not detect these acids. Quantification of these products and the rate constants for the mannitol decomposition network were also obtained. The reaction rate coefficients were determined by fitting the experimental results.

Kenshi Watanabe, Charose Marie Ting Perez, Tomoki Kitahori, Kosuke Hata, Masato Aoi, Hirokazu Takahashi, Tetsushi Sakuma, Yoshiko Okamura, Yutaka Nakashimada, Takashi Yamamoto, Keisuke Matsuyama, Shinzo Mayuzumi, Tsunehiro Aki

Summary: Efficiency of specific gene knock-in by homologous recombination was significantly increased by over 10 times through combining the CRISPR-Cas9 system in Aurantiochytrium strains. Genome edited strains with higher fatty acid productivity were isolated by disrupting genes associated with beta-oxidation, demonstrating the effectiveness of CRISPR-Cas9 for molecular breeding in Aurantiochytrium species to improve lipid productivity.

Summary: Gas fermentation is a promising process for converting CO2 or syngas into valuable chemicals. Homaoacetogens are capable of autotrophic growth using CO2 and H-2 or CO to produce acetate. Through metabolic engineering, it was found that maintaining a level of acetate production along with supplying energy-rich CO allows the homoacetogen to maintain autotrophic growth on syngas.

Summary: Biomineralization is a complex process that controls the deposition of inorganic materials in living organisms with the help of associated proteins. Mms6, a protein isolated from the surfaces of bacterial magnetite nanoparticles, plays a crucial role in regulating the crystal morphology of magnetite. The study demonstrates the direct adsorption ability of Mms6 on magnetite nanoparticles and other inorganic nanoparticles, suggesting its potential use in biomedical and medicinal applications.

Bioaugmentation with marine sediment-derived microbial consortia in mesophilic anaerobic digestion for enhancing methane production under ammonium or salinity stress

Luong Van Duc, Shintaro Nagao, Mohammad Mojarrad, Yuta Miyagawa, Zi-Yan Li, Daisuke Inoue, Takahisa Tajima, Michihiko Ike

Summary: This study evaluated whether bioaugmentation using marine sediment-derived microbial consortia could alleviate the inhibitory effects of ammonium (NH4+) and salinity (NaCl) on methane (CH4) production in anaerobic digestion, and elucidated the underlying mechanisms. The results showed that bioaugmentation enhanced CH4 production compared to non-bioaugmentation. Network analysis revealed the joint effects of microbial connections by Methanoculleus, which promoted the efficient consumption of propionate accumulated under NH4+ and NaCl stresses. Overall, bioaugmentation with pre-acclimated marine sediment-derived microbial consortia can mitigate the inhibition under NH4+ or NaCl stress and enhance CH4 production in anaerobic digestion.

Summary: Thraustochytrid, Aurantiochytrium sp., is a microorganism that produces various useful substances. Lignocellulosic biomass can be used as a feedstock to extract these substances using steam explosion. However, the by-products of steam explosion inhibit the growth and lipid production of Aurantiochytrium sp. This study found that certain by-products were particularly inhibitory, but treating the biomass to reduce the concentration of a major inhibitory component could alleviate the inhibition.
