Long Non-coding RNAs and Their Roles in Non-small-cell Lung Cancer

As a leading cause of cancer deaths worldwide, lung cancer is a collection of diseases with diverse etiologies winch can be broadly classified into small-cell lung cancer (SCLC) and non-small-cell lung cancer (NSCLC). Lung cancer is characterized by genomic and epigenomic alterations; however, mechanisms underlying lung tumorigenesis remain to be elucidated. Long non coding RNAs (lncRNAs) are a group of non-coding RNAs that consist of >200 nucleotides but possess low or no protein-coding potential. Accumulating evidence indicates that abnormal expression of lncRNAs is associated with tumorigenesis of various cancers, including lung cancer, through multiple biological mechanisms involving epigenetic, transcriptional, and post-transcriptional alterations. In this review, we highlight the expression and roles of lncRNAs in NSCLC and discuss their potential clinical applications as diagnostic or prognostic biomarkers, as well as therapeutic targets.

Upregulation of wild-type p53 by small molecule-induced elevation of NQO1 in non-small cell lung cancer cells

Summary: RYL-687 inhibits non-small cell lung cancer cells by upregulating p53, which is achieved by inducing reactive oxygen species and upregulating NQO1 to protect p53 from degradation. Targeting NQO1 is a rational strategy to reactivate p53 and RYL-687 has therapeutic potentials in NSCLCs with wild type p53.

Attention has recently been focused on the ocean as a potential source of novel drugs to fight against malignant neoplasms. Marine compounds have shown promising anticancer activity, but few drugs have been approved for specific cancer treatments by the U.S. Food and Drug Administration.

Innate immune checkpoint Siglec10 in cancers: mining of comprehensive omics data and validation in patient samples

Summary: Siglec10 is a member of innate immune checkpoints that inhibits immune cell activation by interacting with CD24 on tumor cells. Expression of Siglec10 was found to be altered in various cancers, particularly upregulated in kidney renal clear cell carcinoma (KIRC) with negative prognosis correlation. It may play an oncogenic role in KIRC by suppressing immune cell activation and interacting with CD24.

Transcriptional regulation and small compound targeting of ACE2 in lung epithelial cells

Summary: In this study, the researchers evaluated the transcription factors that regulate ACE2 expression and identified 24 potential regulators, with Stat3 being the most significant one. They also found that a compound called 6-OAP could effectively inhibit ACE2 expression and reduce IL-6 production. Moreover, 6-OAP was found to inhibit the entry of SARS-CoV-2 into target cells.

Promising anticancer activities and mechanisms of action of active compounds from the medicinal herb Centipeda minima (L.) A. Braun & Asch

Summary: This review provides a comprehensive summary of the anticancer properties exhibited by active components of Centipeda minima (C. minima). It discusses the anticancer potential of C. minima extracts and the identification of active components. Sesquiterpene lactones, such as 6-O-angeloylplenolin (6-OAP) and arnicolide D, have been found to have similar structures and anticancer mechanisms. Clinical trials have assessed the potential of these components in treating vertex balding, alopecia areata, and glioma. Therefore, a better understanding of the underlying anticancer mechanisms and further clinical trials are important for the development of novel anticancer drugs.

Summary: The natural-product macrolide, 10,11-Dehydrocurvularin (DCV), inhibits NLRP3 inflammasome activation, reducing IL-1 beta secretion and caspase-1 activation without affecting NLRC4 and AIM2 inflammasomes. DCV disrupts the interaction between NEK7 and NLRP3, inhibiting NLRP3 inflammasome activation. Furthermore, DCV ameliorates inflammation in vivo by inhibiting the NLRP3 inflammasome.
