HLA class I variation in Iranian Lur and Kurd populations: high haplotype and allotype diversity with an abundance of KIR ligands

HLA-A, -B and -C alleles of 285 individuals, representing three Iranian Lur populations and one Iranian Kurd population were sequenced completely, yielding human leukocyte antigen (HLA) class I genotypes at high resolution and filling four fields of the official HLA nomenclature. Each population has 87-99 alleles, evenly distributed between the three HLA class I genes, 145 alleles being identified in total. These alleles were already known, named and deposited in the HLA database. The alleles form 316 different HLA A-B-C haplotypes, with each population having between 80 and 112 haplotypes. The four Iranian populations form a related group that is distinguished from other populations, including other Iranians. All four KIR ligands - the A3/11, Bw4, C1 and C2 epitopes - are well represented, particularly Bw4, which is carried by three high-frequency allotypes: HLA-A*24:02, HLA-A*32:01 and HLA-B*51:01. In the Lur and Kurd populations, between 82% and 94% of individuals have the Bw4 epitope, the ligand for KIR3DL1. HLA-B*51:01 is likely of Neandertal origin and associated with Behcet's disease, also known as the Silk Road disease. The Lordegan Lur have the highest frequency of HLA-B*51:01 in the world. This allele is present on 46 Lur and Kurd haplotypes. Present at lower frequency is HLA-B*51:08, which is also associated with Behcet's disease. In the four Iranian populations, 31 haplotypes encode both Bw4(+)HLA-A and Bw4(+)HLA-B, a dual combination of Bw4 epitopes that is relatively rare in other populations, worldwide. This study both demonstrates and emphasizes the value of studying HLA class I polymorphism at highest resolution in anthropologically well-defined populations.

Summary: IgG responses to RBD and NP in COVID-19 patients were found to be dominated by conformational and linear epitopes, respectively. The reactivity of patients' sera with reduced recombinant RBD protein was significantly lower than the native form, while no significant differences were observed for NP protein. The identification of immunodominant epitopes in NP and RBD could be valuable for the development of immunotherapy and diagnostic tools for COVID-19.

The expression pattern of membranous TNF-? is distinct from its intracellular form in breast cancer-draining lymph nodes

Summary: This study focused on the expression of membranous and intracellular forms of TNF-alpha in lymphocytes derived from breast cancer-draining lymph nodes. The results showed that CD4+T cells primarily express the membranous form, while CD8+T cells primarily express the intracellular form. The intensity of membranous TNF-alpha expression in CD19+ cells was correlated with poor prognosticators, and the frequency of membranous TNF-alpha+CD19+ cells showed an inverse correlation with the number of involved lymph nodes.

KIR-HLA gene diversities and susceptibility to lung cancer

Summary: The study reveals the association between KIR and HLA class I ligands with genetic predisposition to lung cancer in the native population of southern Iran. Specific KIR genes and HLA-C1 ligands show significant increase in patients with lung cancer, certain genotypes are positively correlated with disease risk, and iKIRs > aKIRs genotypes are more frequently presented in lung cancer patients.

Proteomics strategies for urothelial bladder cancer diagnosis, prognosis and treatment: Trends for tumor biomarker sources

Summary: Urothelial bladder cancer (UBC) is a complex malignancy with a high recurrence rate. Current diagnostic procedures lack sensitivity and specificity, thus biomarkers have potential value for early diagnosis, prognosis, and immunotherapy. Proteomics methods have rapidly advanced in identifying UBC biomarkers, and a panel of biomarkers may achieve higher diagnostic accuracy. This review evaluates recent literature on UBC proteomics, classifies tumor biomarkers based on their sources, and discusses the limitations and advantages of each source.

The impact of HLA polymorphism on herpesvirus infection and disease

Summary: Human Leukocyte Antigens (HLA) are cell surface molecules that play a central role in coordinating immune responses and undergo strong diversifying natural selection by pathogens. This review synthesizes genetic associations of HLA with herpesvirus infection and disease, identifying HLA alleles associated with diverse herpesvirus-related phenotypes. The study also discusses mechanisms by which different HLA and cognate receptor allotypes direct variable immune responses during herpesvirus infection and pathogenesis.

Summary: The functionality of NK cells is influenced by genetic variation in KIR and HLA genes, which fine-tunes the payload of effector molecules in secretory lysosomes. Granzyme B levels in NK cells are genetically determined by allelic variation in HLA class I genes and serve as a robust metric of NK cell function. Variation in granzyme B levels at rest is closely linked to the killing of target cells.

Production of a biosimilar version of aflibercept to improve VEGF blocker cytotoxicity on endothelial cells

Summary: This study aimed to produce a biosimilar version of aflibercept (AFL) and evaluate its effect when used in combination with other vascular endothelial growth factor (VEGF) blocker drugs. The biosimilar-AFL showed significant inhibition potential on HUVEC cells in a dose-dependent manner. Furthermore, co-treatment with Everolimus (EVR), Lenvatinib (LEN), and Sorafenib (SOR) enhanced the cytotoxicity of biosimilar-AFL on HUVEC cells, with LEN and SOR showing a 10-fold increase in cytotoxicity. The combination of biosimilar-AFL with LEN showed the most efficient result, while the combination with EVR showed the least efficient result. Overall, biosimilar-AFL may improve the efficacy of LEN, EVR, and SOR in reducing the VEGF effect on endothelial cells.

Comparative proteomics analysis in different stages of urothelial bladder cancer for identification of potential biomarkers: highlighted role for antioxidant activity

Summary: The aim of this study is to recognize tumor-associated proteins, which are differentially expressed in different stages of UBC patients compared to non-cancerous tissues. The study found significant changes in protein expression related to oxidative stress and antioxidant activity in UBC patients. These findings may contribute to the development of promising biomarkers for early detection, monitoring, and prognosis of UBC.

Polymorphic KIR3DL3 expression modulates tissue-resident and innate-like T cells

Summary: Most human killer cell immunoglobulin-like receptors (KIR) are expressed by natural killer (NK) cells and recognize HLA class I molecules as ligands. However, KIR3DL3 is expressed in γδ and CD8(+) T cells and recognizes the B7 family ligand HHLA2, playing a role in immune checkpoint targeting. KIR3DL3(+) T cells in peripheral blood have an activated transitional memory phenotype and are hypofunctional, with biased T cell receptor (TCR) usage. Additionally, KIR3DL3 expression can be influenced by polymorphisms and variants in the proximal promoter and residue 86. These findings have implications for personalized targeting of KIR3DL3/HHLA2 checkpoint inhibition.

Epidemiological Profile of Salivary Gland Tumors in Southern Iranian Population: A Retrospective Study of 405 Cases
