Serum Brain-Derived Neurotrophic Factor is Related to Platelet Reactivity but not to Genetic Polymorphisms within BDNF Encoding Gene in Patients with Type 2 Diabetes

Background: The aim of this study was to investigate the association between serum concentrations of the brain-derived neurotrophic factor (BDNF), platelet reactivity and inflammatory markers, as well as its association with BDNF encoding gene variants in type 2 diabetic patients (T2DM) during acetylsalicylic acid (ASA) therapy. Material/Methods: This retrospective, open-label study enrolled 91 patients. Serum BDNF, genotype variants, hematological, biochemical, and inflammatory markers were measured. Blood samples were taken in the morning 2-3 h after the last ASA dose. The BDNF genotypes for selected variants were analyzed by use of the iPLEX Sequenom assay. Results: In multivariate linear regression analysis, CADP-CT >74 sec (p<0.001) and sP-selectin concentration (p=0.03) were predictive of high serum BDNF. In multivariate logistic regression analysis, CADP-CT >74 sec (p=0.02) and IL-6 concentration (p=0.03) were risk factors for serum BDNF above the median. Non-significant differences were observed between intronic SNP rs925946, missense SNP rs6265, and intronic SNP rs4923463 allelic groups and BDNF concentrations in the investigated cohort. Conclusions: Chronic inflammatory condition and enhanced immune system are associated with the production of BDNF, which may be why the serum BDNF level in T2DM patients with high platelet reactivity was higher compared to subjects with normal platelet reactivity in this study.

External validation of VO2max prediction models based on recreational and elite endurance athletes

Summary: This study compared predicted VO2max (pVO2max) with directly measured VO2max and found that the accuracy of available prediction models has not been validated in endurance athletes. The models underestimated pVO2max in younger and higher trained athletes, and overestimated pVO2max in older and lower trained athletes. These models should only be used as a supplemental method for physicians and new models need to be developed for clinical practice and sports diagnostics.

The findings suggest that increased levels of trimethylamine-N-oxide (TMAO) may be an independent predictor of MACCE and contribute to post-AMI cardiac dysfunction.

Serum levels of matrix metalloproteinase 2 and its inhibitor after tonic-clonic seizures

Summary: Damage to the blood-brain barrier (BBB) from neuroinflammation after epileptic seizures can disrupt brain homeostasis. This study examined the involvement of modulatory proteins in BBB integration. Serum analysis of 50 epilepsy patients showed slight increases in MMP-2 levels after seizures, along with decreases in TSP-1 and -2 levels and an increase in TIMP-2 expression. However, there were no significant changes in alpha 2-macroglobulin and MMP-7 levels. These findings suggest the initiation of repair processes and improved integrity of the BBB after seizures. Further studies correlating serum markers with brain expression are needed for potential identification of epileptic seizure biomarkers.

Olfactory dysfunction in patients with Wilson?s Disease

Summary: This study investigated the relationship between blood-derived microRNAs (miRNAs) and long-term all-cause mortality in patients with non-ST-segment elevation acute coronary syndrome (NSTE-ACS). The expression of miR-223 was found to be associated with improved long-term survival in NSTE-ACS patients. This hypothesis-generating study suggests that miR-223 may be a suitable predictor for long-term all-cause mortality.

Inflammatory Forms of Cardiomyocyte Cell Death in the Rat Model of Isoprenaline-Induced Takotsubo Syndrome

Summary: The aim of this study was to evaluate the presence and dynamics of necroptosis and pyroptosis in the rat model of TTS. The results showed higher plasma concentrations of interleukin-1β in TTS patients compared to controls. Protein expression associated with necroptosis and pyroptosis was observed in the myocardium after the injection of isoprenaline. These findings suggest that TTS is associated with inflammatory-associated programmed cell death and inflammation may occur simultaneously with cardiomyocyte death.
