Multi-walled carbon nanotubes increase antibody-producing B cells in mice immunized with a tetravalent vaccine candidate for dengue virus

Background: In recent times, studies have demonstrated that carbon nanotubes are good candidates for use as vehicles for transfection of exogenous material into the cells. However, there are few studies evaluating the behavior of carbon nanotubes as DNA vectors and few of these studies have used multi-walled carbon nanotubes (MWCNTs) or carboxylated MWCNTs. Thus, this study aims to assess the MWCNTs' (carboxylated or not) efficiency in the increase in expression of the tetravalent vaccine candidate (TVC) plasmid vector for dengue virus in vitro using Vero cells, and in vivo, through the intramuscular route, to evaluate the immunological response profile. Results: Multi-walled carbon nanotubes internalized by Vero cells, have been found in the cytoplasm and nucleus associated with the plasmid. However, it was not efficient to increase the messenger ribonucleic acid (mRNA) compared to the pure vaccine candidate associated with Lipofectamine (R) 2000. The in vivo experiments showed that the use of intramuscular injection of the TVC in combination with MWCNTs reduced the immune response compared to pure TVC, in a general way, although an increase was observed in the population of the antibody-producing B cells, as compared to pure TVC. Conclusions: The results confirm the data found by other authors, which demonstrate the ability of nanotubes to penetrate target cells and reach both the cytoplasm and the cell nucleus. The cytotoxicity values are also in accordance with the literature, which range from 5 to 20 mu g/mL. This has been found to be 10 mu g/mL in this study. Although the expression levels are higher in cells that receive the pure TVC transfected using Lipofectamine (R) 2000, the nanotubes show an increase in B-cells producing antibodies.

A Rapid Method for Performing a Multivariate Optimization of Phage Production Using the RCCD Approach

Summary: This study evaluated the production of a T4-like bacteriophage in seven types of culture media and optimized the process by using four cultivation variables. The rotatable central composite design methodology was used to determine the ideal conditions for scaling up production. Despite the high number of conditions evaluated, the results showed that effective viral production could be achieved even with minimal medium, reducing costs during large-scale production.

Computational Guided Method Applied to LSPR-Based Biosensor for Specific Detection of the Four-Serotypes of Dengue Virus in Seropositive Patients

Summary: This paper presents a localized surface plasmon resonance (LSPR)-based biosensor for selectively detecting four serotypes of Dengue virus (DENVx) in seropositive patients. The behavior of gold nanoparticles in the shape of nanorods is studied theoretically and numerically, with induced structural variations observed during the detection process. The predictability of experimental results is calculated using the largest Lyapunov exponent, revealing chaotic systems with low repetition probability. The genetic similarity between certain DENV serotypes is evidenced through analysis of the resonance curve generated by the LSPR system. The biosensors are validated using samples from seronegative patients for DENVx and seropositive patients for other Flaviviruses.

Summary: This study aimed to identify MHC-I-restricted peptide signatures for arboviruses using in silico and in vitro peptide microarray tools. The results showed that there were few overlapping immunogenic peptides between different arboviruses, with DENV-derived peptides showing the highest reactivity with HLA-A*02:01. Additionally, non-structural proteins were identified as hotspots for HLA-A*02:01-reactive peptides.

Summary: This study evaluated the performance of the VIDAS(R) dengue immunoassays in diagnosing dengue virus infections. The results showed that these immunoassays have high accuracy and stability in detecting NS1 antigen and anti-DENV IgM and IgG antibodies. They are suitable for the accurate diagnosis of dengue infections in endemic regions.

Summary: Proteus mirabilis is a significant pathogen causing catheter-associated urinary tract infections (CAUTIs). Two phages, BigMira and MidiMira, were studied and found to have similarities with four missense mutations impacting their depolymerase activity. Analysis of clinical P. mirabilis strains revealed differences in antibiotic resistance profiles and lipopolysaccharide locus, explaining phages' host range. The presence of antimicrobial resistance genes poses challenges for treating infections caused by multidrug-resistant bacteria.

Summary: The emergence of the Brazilian Gamma variant has impacted the epidemiological profile of COVID-19 cases, with higher transmissibility and immune evasion ability. The study showed that the Gamma variant rapidly increased in prevalence two months after its first detection in Sao Paulo, Brazil, and displaced seven circulating variants. Additionally, it was associated with increased mortality risk and severity of COVID-19 cases in younger age groups.
