Summary: Real-world biomarker assessments in adult SCD patients at the University of Illinois revealed significant differences in markers of haemolysis, inflammation, and organ function between severe and mild genotypes. While untreated mild genotype patients showed biomarkers closer to control ranges, severe genotype patients receiving treatment did not significantly improve, indicating a need for more effective treatments.

Summary: The aberrant proliferation of pulmonary artery smooth muscle cells caused by over-expression of SPHK1 leads to changes in genes regulating cellular immune response, with elevated levels of IL6, STAT1, and PARP9. These differentially expressed genes are mapped to immune-modulatory signaling networks, including IFNG. Additionally, PARP9 and STAT1 proteins were found to be elevated in primary hPASMCs isolated from PAH patients.

Elevated LH/FSH ratio and free testosterone levels were observed in both adult and adolescent PCOS patients compared to controls. Genotype-Guided vs Clinically-Guided Stable Warfarin Dose Prediction and Stable Dose Establishment In A Predominantly Non-European Ancestry Population

Summary: Genotype-guided warfarin dosing did not improve eventual stable dose discordance or reduce the time to achieve stable dose in this predominantly non-European cohort. Dietary vitamin K intake level did not impact warfarin dose discordance or time to achieve stable warfarin dose. Additional study is needed to identify populations that best benefit from PGx-guided warfarin dosing.

Cortactin loss protects against hemin-induced acute lung injury in sickle cell disease

Summary: This study investigated the role of the cytoskeletal protein cortactin (CTTN) in hemin-induced acute lung injury (ALI) and acute chest syndrome (ACS). The results showed that cortactin deficiency provided protection against hemin-induced injury in mice and in vitro cell models, suggesting a new potential treatment avenue for this devastating disease.
