Inflammatory airway diseases are a significant health problems requiring new approaches to the existing therapies and addressing fundamental issues. Difficulties in developing effective therapeutic strategies might be caused by lack of understanding of their exact molecular mechanism. MicroRNAs (miRNAs) are a class of regulators that already revolutionized the view of gene expression regulation. A cumulating number of investigations show a pivotal role of miRNAs in the pathogenesis of asthma, chronic obstructive pulmonary disease (COPD), or airway remodeling through the regulation of many pathways involved in their pathogenesis. Expression changes of several miRNAs have also been found to play a role in the development and/or improvement in asthma or COPD. Still, relatively little is known about the role of miRNAs in inflammatory disorders. The microRNA profiles may differ depending on the cell type or antigen-presenting cell. Based on the newest literature, this review discusses the current knowledge concerning miRNA contribution and influence on lung inflammation and chosen inflammatory airway diseases: asthma and COPD.

Curcumin modulates airway remodelling-contributing genes-the significance of transcription factors

Joanna Wieczfinska, Przemyslaw Sitarek, Tomasz Kowalczyk, Piotr Rieske, Rafal Pawliczak

Summary: Bronchial epithelial cells and fibroblasts are crucial in airway remodelling, with human rhinovirus infection playing a role in this process. Curcumin has been shown to reduce gene expression related to airway remodelling and may potentially limit or prevent this process, dependent on NF-kappa B, c-Myc, and STAT3 pathways. Further research is needed to fully understand the effects of curcumin on airway remodelling.

Genetic Manipulation and Bioreactor Culture of Plants as a Tool for Industry and Its Applications

Tomasz Kowalczyk, Anna Merecz-Sadowska, Laurent Picot, Irena Brcic Karaconji, Joanna Wieczfinska, Tomasz Sliwinski, Przemyslaw Sitarek

Summary: In recent years, there has been growing interest in using transgenic plants as biofactories for the production of valuable secondary metabolites and recombinant proteins. Advances in genetic engineering technology have allowed for direct modification of genes involved in the biosynthesis of these compounds. Research has resulted in efficient plant systems that can produce specific metabolites and proteins, and the use of bioreactors enables industrial-scale production. This review paper provides an overview of the key research in the past decade, highlighting the use of transgenic plants as small, green biofactories and discussing the current state of patents in this field.

Oxidative Stress-Related Mechanisms in SARS-CoV-2 Infections

Joanna Wieczfinska, Paulina Kleniewska, Rafal Pawliczak

Summary: The COVID-19 pandemic has resulted in high mortality, particularly in patients with comorbidities such as diabetes, hypertension, and COPD. Oxidative stress is believed to play a significant role in the pathogenesis of these comorbidities. Although the exact mechanism of death in critically ill COVID-19 patients is still unclear, lung vasculature injury and loss of lung function are considered crucial. Medications used in COVID-19 treatment that influence reactive oxygen and nitrogen species production may be beneficial. However, oxidative stress is also involved in antiviral immune response, and understanding the redox mechanisms in severe COVID-19 could impact its treatment.

Comparison of effects of tobacco cigarettes, electronic nicotine delivery systems and tobacco heating products on miRNA-mediated gene expression. A systematic review

Paulina Natalia Kopa-Stojak, Rafal Pawliczak

Summary: This study aims to summarize the current knowledge on the effects of cigarettes, electronic nicotine delivery systems, and tobacco heating products on miRNA-mediated gene expression regulation and their potential impact on smoking-related respiratory disease development. A literature search was conducted from October 2021 to February 2022, including relevant literature such as clinical trials, cellular and animal-based studies. The results showed that cigarette smoke significantly altered the transcriptome and miRNA expression profile. Although the effects of electronic cigarettes and tobacco heating products on miRNA profile were reduced compared to cigarette smoke, there were still significant differences in miRNA expression compared to air-control, which may be harmful to never-smokers.

COVID-19 vaccination efficacy in numbers including SARS-CoV-2 variants and age comparison: a meta-analysis of randomized clinical trials

Marharyta Sobczak, Rafal Pawliczak

Summary: This study compared the efficacy of COVID-19 vaccines and found that full vaccination can decrease the risk of infection and death. The vaccines are also effective against variants of SARS-CoV-2. The efficacy of vaccination is similar in different age groups.

Factors That Affect the COVID-19 Pandemic in Summer 2022 Compared to Summer 2021

Marharyta Sobczak, Rafal Pawliczak

Summary: This study compares the factors contributing to the increasing number of COVID-19 cases in five selected European Union countries in 2021 and 2022. The research findings indicate that despite high vaccination rates, these countries experienced a sharp increase in daily cases during the summer of 2022, potentially due to the presence of the Omicron variant.

COVID-19 mortality rate determinants in selected Eastern European countries

Marharyta Sobczak, Rafal Pawliczak

Summary: This study aimed to investigate the common factors responsible for the increased COVID-19 mortality in Eastern European countries. The findings showed that COVID-19 mortality is associated with the number of cases, hospitalizations, and patients in intensive care. Stringency index and vaccination also have an impact on COVID-19 mortality. Moreover, the presence of new variants increased the death rates.

Summary: Accumulating evidence suggests that metformin has anticancer properties by inducing apoptosis and inhibiting the growth and proliferation of cancer cells. However, its effect on histone acetylation status remains unclear. Therefore, this study aimed to evaluate the impact of metformin on HATs and HDACs in human pancreatic cancer cell lines.
