Facile preparation of salivary extracellular vesicles for cancer proteomics

Extracellular vesicles (EVs) are membrane surrounded structures released by cells, which have been increasingly recognized as mediators of intercellular communication. Recent reports indicate that EVs participate in important biological processes and could serve as potential source for cancer biomarkers. As an attractive EVs source with merit of non-invasiveness, human saliva is a unique medium for clinical diagnostics. Thus, we proposed a facile approach to prepare salivary extracellular vesicles (SEVs). Affinity chromatography column combined with filter system (ACCF) was developed to efficiently remove the high abundant proteins and viscous interferences of saliva. Protein profiling in the SEVs obtained by this strategy was compared with conventional centrifugation method, which demonstrated that about 70% more SEVs proteins could be revealed. To explore its utility for cancer proteomics, we analyzed the proteome of SEVs in lung cancer patients and normal controls. Shotgun proteomic analysis illustrated that 113 and 95 proteins have been identified in cancer group and control group, respectively. Among those 63 proteins that have been consistently discovered only in cancer group, 12 proteins are lung cancer related. Our results demonstrated that SEVs prepared through the developed strategy are valuable samples for proteomics and could serve as a promising liquid biopsy for cancer.

Ti4+-IMAC was shown to be the most efficient method for phosphopeptide isolation from sEVs. By using this approach, over 500 sEV phosphopeptides were identified, providing a comprehensive analysis for the detection of lung cancer non-invasively.

Phosphoproteomics of extracellular vesicles integrated with multiomics analysis reveals novel kinase networks for lung cancer

In-Depth Metaproteomics Analysis of Oral Microbiome for Lung Cancer

Quadruple UV LED Array for Facile, Portable, and Online Intrinsic Fluorescent Imaging of Protein in a Whole Gel Electrophoresis Chip

Summary: In this study, a quadruple UV LED array was designed to directly irradiate gel electrophoresis chips at 275 nm. Simple detection setup using a 365 nm filter, UV camera lens, and CCD was integrated with the small gel electrophoresis chip to construct the IFI-GEC device. The LED array exhibited good evenness and enabled online imaging of the entire gel electrophoresis chip. The reproducible IFI-GEC detection was completed within 10 minutes with a low LOD of 40 ng for lysozyme detection, demonstrating the potential of the IFI-GEC device for stain-free online and portable protein detection.

Isoelectric point barcode and similarity analysis with the earth mover's distance for identification of species origin of raw meat

Summary: In this study, we introduced the concept of isoelectric point (pI) barcode for species identification of raw meat by combining microcolumn isoelectric focusing (mIEF) and similarity analysis with the earth mover's distance (EMD) metric. We analyzed 14 meat species using mIEF to generate electropherograms of myoglobin/hemoglobin (Mb/Hb) markers, which were then converted into pI barcodes for EMD analysis. By developing a barcode database and using the EMD method, we successfully identified 9 meat products with the advantages of convenience, speed, and low cost.

Discovery of Dimethyl Shikonin Oxime 5a, a Potent, Selective Bombesin Receptor Subtype-3 Agonist for the Treatment of Type 2 Diabetes Mellitus

Summary: The study discovered the close association between DMAKO-00 and its derivatives with BB3, with DSO-5a identified as the most potent agonist for BB3. DSO-5a improved glucose tolerance in C57BL/6 mice and glucose homeostasis in diabetic db/db mice through BB3. It was further revealed that DSO-5a upregulated PPAR-γ activity via BB3 through quantitative proteomics. Overall, the study demonstrated that DSO-5a is a potent, selective, and low-brain-penetrating agonist for BB3, and BB3 is a promising treatment target for type 2 diabetes mellitus.
