Reducing central serotonin in adulthood promotes hippocampal neurogenesis

Chronic administration of selective serotonin reuptake inhibitors (SSRIs), which up-regulates central serotonin (5-HT) system function, enhances adult hippocampal neurogenesis. However, the relationship between central 5-HT system and adult neurogenesis has not fully been understood. Here, we report that lowering 5-HT level in adulthood is also able to enhance adult hippocampal neurogenesis. We used tamoxifen (TM)-induced Cre in Pet1-CreERT2 mice to either deplete central serotonergic (5-HTergic) neurons or inactivate 5-HT synthesis in adulthood and explore the role of central 5-HT in adult hippocampal neurogenesis. A dramatic increase in hippocampal neurogenesis is present in these two central 5-HT-deficient mice and it is largely prevented by administration of agonist for 5-HTR2c receptor. In addition, the survival of new-born neurons in the hippocampus is enhanced. Furthermore, the adult 5-HT-deficient mice showed reduced depression-like behaviors but enhanced contextual fear memory. These findings demonstrate that lowering central 5-HT function in adulthood can also enhance adult hippocampal neurogenesis, thus revealing a new aspect of central 5-HT in regulating adult neurogenesis.

Summary: This study revealed the regulatory role of CDH13 in inhibitory synapse function by establishing a human neuronal model. By culturing iGABAs in vitro, we found that CDH13 deficiency increases inhibition, thereby reducing the excitatory/inhibitory balance. Through these findings, we identified a potential mechanism by which CDH13 may contribute to the pathophysiology of brain disorders.

Satb2 Regulates EphA7 to Control Soma Spacing and Self-Avoidance of Cortical Pyramidal Neurons

Summary: Soma spacing and dendritic arborization are crucial for the establishment of proper neural circuitry and function during brain development. This study reveals the involvement of transcription factor Satb2 in regulating these processes. Satb2 is shown to be necessary for the regionalization of retrosplenial cortex, the determination of callosal neuron fate, and the regulation of soma spacing and dendritic self-avoidance of cortical pyramidal neurons. The study also identifies EphA7 as a downstream effector of Satb2, with Satb2 directly suppressing the expression of EphA7. These findings provide new insights into the transcriptional regulation of cerebral cortex morphogenesis.

However, a single dimension of genetic risk does not appear to be useful in explaining the biobehavioral correlates or individual variants of psychiatric disorders.

High Morphine Use Disorder Susceptibility Is Predicted by Impaired Learning Ability in Mice

Summary: This study found that learning ability in mice is negatively correlated with susceptibility to morphine use disorder (MUD), suggesting that impaired learning ability can predict SUD susceptibility. FVB mice, which showed impaired learning ability, exhibited higher sensitivity to MUDs compared to C57 mice. These findings highlight the importance of considering learning ability in identifying high-risk SUD groups.

A morphine reward generalization mouse model based on conditioned place preference and aversion

Summary: In this study, a simple and effective experimental model was designed to investigate the generalization memory of morphine reward. By replacing the conditioning CPP box, it was found that mice still showed a clear preference for generalized information. The establishment of this study provides a new tool for studying generalization memory in substance use disorders.
