Oxidative polymerization of tyrosol by horseradish peroxidase (HRP)-H2O2 afforded an insoluble product (oligotyrosol, OligoTyr) consisting of mixture of linear oligomers (up to 11-mer) with limited benzylic branching points, as evidenced by ESI-MS and solid state C-13 NMR analysis. OligoTyr proved to be significantly more active than tyrosol in several antioxidant assays and was not toxic to human osteosarcoma SaOS-2 cells, stimulating alkaline phosphatase (ALP) activity at day 7 in a similar manner as tyrosol. However, when loaded at 5% w/w into highly porous polylactic acid (PLA) scaffolds featuring hierarchical structures, OligoTyr caused a significant increase in the ALP activity of SaOS-2 cells compared to PLA alone, while tyrosol was completely inactive. A release of ca. 5% from PLA was determined after 1 week in a physiological medium. No significant influence on calcium release from PLA scaffolds containing 5% beta-tricalcium phosphate was observed.

Endoplasmic Reticulum Stress and Cancer: Could Unfolded Protein Response Be a Druggable Target for Cancer Therapy?

Gregorio Bonsignore, Simona Martinotti, Elia Ranzato

Summary: The unfolded protein response (UPR) is an adaptive response triggered by endoplasmic reticulum (ER) stress for re-establishing protein homeostasis. Specific ER proteins mediate UPR activation by dissociating from chaperone Glucose-Regulated Protein 78 (GRP78). UPR can decrease ER stress, produce an ER adaptive response, block UPR if ER homeostasis is restored, or regulate apoptosis. This review focuses on molecules that target ER stress and demonstrate how UPR could be a new target in cancer treatment.

The Effect of Cosmetic Ingredients of Phenol Type on Immediate Pigment Darkening and Their (Photo)Protective Action in Association with Melanin Pigmentation: A Model In Vitro Study

Sara Viggiano, Lucia Panzella, Maria Reichenbach, Joachim Hans, Alessandra Napolitano

Summary: Immediate pigment darkening, the rapid onset of melanin pigmentation resulting from oxidation, can lead to irregular pigmentation and dark spots. Phenolic cosmetic ingredients (CIs) such as paradol-6 and phenylethyl resorcinol can decrease the oxidation and melanin formation of 5,6-dihydroxyindole (DHI) and 5,6-dihydroxyindole-2-carboxylic acid (DHICA). CIs also show photoprotective effects in model systems, reducing peroxidation and generation of reactive oxygen species. These findings have potential implications for the development of skincare formulations to improve skin spots and counteract aging processes associated with sun exposure.

Rita Argenziano, Maria Laura Alfieri, Noemi Gallucci, Gerardino D'Errico, Lucia Panzella, Alessandra Napolitano

Summary: The search for new synthetic melanin-related pigments with antioxidant and photoprotective properties is important for dermo-cosmetic purposes. In this study, a melanin derived from 5,6-dihydroxyindole-2-carboxylic acid (DHICA) was synthesized and found to have similar structural properties to DHICA melanin. The synthesized pigment demonstrated stronger UV-visible absorption and improved solubility compared to DHICA melanin. Furthermore, the pigment exhibited marked antioxidant properties and enhanced inhibitory action against lipid peroxidation. These findings suggest that the melanin derived from DHICA has potential as a functional ingredient in dermo-cosmetic formulations.

Biosourced Aromatic Derivatives in the Upcycling of Recycled PET: Mellophanic Dianhydride as a Chain Extender

Ada Truscello, Sonia Colletti, Cristian Gambarotti, Marco A. Ortenzi, Stefano Gazzotti, Maurizio S. Galimberti

Summary: This study reports the synthesis of biosourced mellophanic dianhydride (MEDA) from agricultural waste and its use as a chain extender for mechanically recycled PET (R-PET) as a sustainable alternative to oil-based pyromellitic dianhydride (PMDA). The rheological tests showed similar results between R-PET extruded with MEDA and PMDA, and DMTA analysis indicated that R-PET containing MEDA had a 20% higher Young's modulus compared to pristine R-PET in the temperature range commonly used for blow molding bottles. The advantage of MEDA lies in its sustainable production using agricultural waste, while PMDA is derived from oil-based chemistry.

Performance comparison of coatings based on cellulose nanocrystals and microfibrillated cellulose for food packaging

Ghislain Fotie, Stefano Gazzotti, Marco Aldo Ortenzi, Sara Limbo, Luciano Piergiovanni

Summary: This article compares the gas barrier performance of plastic films coated with cellulose nanocrystals (CNCs) and microfibrillated cellulose (MFC) obtained from cotton linters. The CNCs were chemically isolated using ammonium persulfate (APS) hydrolysis, while the MFC was obtained mechanically either with or without APS-assisted pre-treatment. Mechanical tests and various characterization techniques were performed on the samples. Coated PET films were then evaluated for water contact angle, transparency, opacity, water vapor transmission rate, and oxygen transmission rate. The findings show that MFCaps-based coated films have better oxygen barrier properties at higher relative humidity and higher residue levels during degradation compared to CNCs.
