摘要

Bone morphogenetic proteins (BMPs) play important roles in embryonic and postnatal development by regulating cell differentiation, proliferation, motility, and survival, thus maintaining homeostasis during organ and tissue development. BMPs can lead to tumorigenesis and regulate cancer progression in different stages. Therefore, we summarized studies on BMP expression, the clinical significance of BMP dysfunction in various cancer types, and the molecular regulation of various BMP-related signaling pathways. We emphasized on the paradoxical effects of BMPs on various aspects of carcinogenesis, including epithelial-mesenchymal transition (EMT), cancer stem cells (CSCs), and angiogenesis. We also reviewed the molecular mechanisms by which BMPs regulate tumor generation and progression as well as potential therapeutic targets against BMPs that might be valuable in preventing tumor growth and invasion.

Tetrandrine Inhibits Cancer Stem Cell Characteristics and Epithelial to Mesenchymal Transition in Triple-Negative Breast Cancer via SOD1/ROS Signaling Pathway

Summary: In this study, the efficacy of tetrandrine in targeting cancer stemness and epithelial to mesenchymal transition (EMT) in triple-negative breast cancer (TNBC) was evaluated. Tetrandrine exhibited antiproliferative and cell cycle arrest activities, reduced cancer stem cell population, prevented mammosphere formation, and suppressed cell migration and invasion. It was found that tetrandrine downregulated SOD1 and catalase expression, induced ROS production, and inhibited EMT and stemness in TNBC cells.

Mechanistically, NCAPG activates the Wnt/beta-catenin signaling pathway to facilitate CRC invasion and metastasis.

Summary: ARHGAP10 was found to have lower expression in NSCLC tissues compared with normal tissues, and overexpression of ARHGAP10 inhibited the migratory and invasive potential of lung cancer cells by suppressing EMT through the PI3K/Akt/GSK3 beta pathway. Activation of the IGF-1 signaling pathway can reverse the effects of ARHGAP10 on EMT regulation in NSCLC cells.

Epithelial membrane protein 3 regulates lung cancer stem cells via the TGF-β signaling pathway

Summary: EMP3 is a transmembrane glycoprotein initially thought to be a tumor suppressor, but it has been found to potentially promote tumor growth. In non-small cell lung cancer cells overexpressing ALDH1, EMP3 is upregulated and involved in cell proliferation, CSCs formation, and EMT. It interacts with TGFBR2, regulating TGF-beta/Smad signaling and affecting CSCs and EMT. High EMP3 expression is associated with poor patient survival rates.

Summary: In this study, the upregulation of ACTN1 in gastric cancer was associated with poor prognosis, and over-expression of ACTN1 promoted proliferation, invasion, and migration of GC cells. This process was mediated through the AKT/GSK3β/β-catenin signaling pathway and confirmed by the AKT inhibitor MK2206. These findings suggest that ACTN1 may be a promising candidate for gastric cancer treatment.
