Summary: In this study, the efficacy of tetrandrine in targeting cancer stemness and epithelial to mesenchymal transition (EMT) in triple-negative breast cancer (TNBC) was evaluated. Tetrandrine exhibited antiproliferative and cell cycle arrest activities, reduced cancer stem cell population, prevented mammosphere formation, and suppressed cell migration and invasion. It was found that tetrandrine downregulated SOD1 and catalase expression, induced ROS production, and inhibited EMT and stemness in TNBC cells.

Summary: The polysaccharides of Ganoderma lucidum have the potential to be an important ally for tongue squamous cell carcinoma treatment, as they can alter the behavior and morphology of tumor cells, as well as downregulate cancer stem cells and EMT markers.

Summary: This study reveals the crucial role of TWIST1 in the establishment of a cancer stem cell-like phenotype, EMT progression, and inhibition of apoptosis in esophageal squamous cell carcinoma (ESCC). These findings provide evidence for TWIST1 as a therapeutic target for suppressing CSC generation and ESCC malignancy.

Summary: The epithelial to mesenchymal transition (EMT) is a cellular process crucial in embryogenesis, where cancer stem cells (CSCs) play a role by differentiating and undergoing EMT. Long noncoding RNAs (lncRNAs) also have a role in CSCs and EMT, with the potential to serve as biomarkers and therapeutic targets in cancer progression.

Summary: NCAPG is over-expressed in colorectal cancer and is associated with clinicopathological characteristics. Silencing NCAPG can suppress the proliferation, migration, and invasion of CRC cells, while overexpression of NCAPG promotes these abilities. Mechanistically, NCAPG activates the Wnt/beta-catenin signaling pathway to facilitate CRC invasion and metastasis.

Summary: ARHGAP10 was found to have lower expression in NSCLC tissues compared with normal tissues, and overexpression of ARHGAP10 inhibited the migratory and invasive potential of lung cancer cells by suppressing EMT through the PI3K/Akt/GSK3 beta pathway. Activation of the IGF-1 signaling pathway can reverse the effects of ARHGAP10 on EMT regulation in NSCLC cells.

Summary: EMP3 is a transmembrane glycoprotein initially thought to be a tumor suppressor, but it has been found to potentially promote tumor growth. In non-small cell lung cancer cells overexpressing ALDH1, EMP3 is upregulated and involved in cell proliferation, CSCs formation, and EMT. It interacts with TGFBR2, regulating TGF-beta/Smad signaling and affecting CSCs and EMT. High EMP3 expression is associated with poor patient survival rates.

Summary: The study revealed that MSCs from lung cancer promoted tumor metastasis and tumorigenesis by inducing partial EMT and upregulating stem cell-related genes in lung cancer cells. These findings suggest that LC-MSCs may serve as potential targets or vehicles for lung cancer therapy.

Summary: In this study, the upregulation of ACTN1 in gastric cancer was associated with poor prognosis, and over-expression of ACTN1 promoted proliferation, invasion, and migration of GC cells. This process was mediated through the AKT/GSK3β/β-catenin signaling pathway and confirmed by the AKT inhibitor MK2206. These findings suggest that ACTN1 may be a promising candidate for gastric cancer treatment.

Summary: The interplay between epithelial-mesenchymal plasticity and cancer stem cells plays a crucial role in cancer progression, affecting the aggressiveness of tumor cells and their response to treatment. Understanding the dynamic nature of EMT as a hybrid state rather than a binary model is important for unraveling the complexities of cancer biology and developing targeted therapeutic interventions.

Summary: In this study, a novel signaling axis mediated by KMT2D linking TGF-beta to the activin A pathway was discovered. Loss of KMT2D induces the expression and secretion of activin A, which activates a noncanonical p38 MAPK-mediated pathway to enhance tumor invasion and metastasis in pancreatic cancer.
