Summary: The peroxidase family of peroxiredoxins (PRDXs) plays a vital role in maintaining the intracellular balance of ROS in head and neck squamous cell carcinoma (HNSCC). The expression of PRDX1, PRDX4, and PRDX5 in HNSCC is increased, while the expression of PRDX2 is decreased. Higher expression of PRDX4/5/6 indicates poor prognosis, while lower expression of PRDX1/5 is associated with more immune cell infiltration, higher expression of immune-related molecules, and a likely response to anti-PD-1 treatment. Knockdown of PRDX5 inhibits HNSCC cell proliferation, invasion, and metastasis, and promotes apoptosis through its antioxidant property. This study highlights the potential role of PRDXs, particularly PRDX5, in HNSCC and suggests it as a potential therapeutic target.

Summary: This study investigated the effect of Notch2 gene knockout in Treg cells on head and neck squamous cell carcinoma (HNSCC) in mice. The results showed that specific knockout of the Notch2 gene in Treg cells significantly decreased tumor volume, reduced the number of Treg cells, inhibited tumor growth, and improved the immune microenvironment in mice.

Summary: In this study, a metabolism-related gene prognostic index (MRGPI) for head and neck squamous cell carcinoma (HNSCC) was constructed, which bridged metabolic characteristics and antitumor immune cycling, and the immunophenotype, genetic alterations, potential targeted inhibitors, and the benefit of immunotherapy were identified in MRGPI-defined HNSCC subgroups. Patients with a low MRGPI score displayed lower metabolic activities and an active antitumor immunity status and had more benefit from immunotherapy, while a higher MRGPI score was associated with higher metabolic activities, lower antitumor immunity ability, an immunosuppressive tumor microenvironment, and less benefit from immunotherapy.

Summary: This study aimed to construct a risk model based on therapeutic response-related genes (TRRGs) to predict therapy response and prognosis in TRRGs-defined subgroups of head and neck squamous cell carcinoma (HNSCC). By analyzing data from TCGA and GEO databases, differentially expressed TRRGs significantly associated with survival were identified, and a signature and prognostic nomogram based on TRRGs were established. The results showed that the model could effectively predict therapy response and overall survival in HNSCC patients.
