Summary: This study found that supplementation with a Lactobacillus species producing feruloyl esterase (FAEb) can alleviate ulcerative colitis in mice by reducing body weight loss, colon length shortening, fecal occult blood severity, and disease activity index. FAEb supplementation also improves antioxidant capacity, promotes beneficial short-chain fatty acid production, increases expression of tight junction proteins, reduces histopathological scores, and prevents mucous barrier damage in the gut. Additionally, FAEb supplementation inhibits inflammatory signaling pathways, increases beneficial bacteria abundance, and regulates gut microbiota balance.

Summary: The study found that esterases from various bacteria and fungi can catalyze the release of diferulic acids from complex arabinoxylans. By comparing the activity of the enzymes on synthetic substrates and diFA release, a detailed structure-function analysis is needed to explain the diFA releasing ability of the enzymes. The findings contribute to a better understanding of the role of esterases in the degradation of complex arabinoxylans and provide new opportunities for enzyme-assisted upgrading of complex bran arabinoxylans.

Summary: In this study, a feruloyl esterase (LaFae) from Lactobacillus acidophilus was obtained and characterized. LaFae could effectively produce ferulic acid from wheat bran, rice bran, and corn stalks. Xylanase supplementation was found to enhance LaFae activity, increasing ferulic acid production. The crystal structures of LaFae revealed the active site configuration and the role of a gatekeeper residue in substrate binding.

Summary: The metabolism of hydroxybenzoic and hydroxycinnamic acid derivatives in Lactobacillus plantarum involves a two-step pathway including esterase action and decarboxylation. Cloned esterase genes from L. plantarum were introduced into lactic acid bacteria to increase the bioavailability of healthy compounds in food. This study demonstrated the potential for utilizing these genes to enhance the metabolic pathways of phenolic acids in relevant lactic acid bacteria.

Summary: A novel esterase gene was isolated from a soil metagenomic library, which exhibited high activity towards p-nitrophenyl esters with short acyl chains and p-nitrophenyl acetate. The enzyme showed optimal activity at pH 7.0 and 30 degrees C, and was stable in a broad pH range from 4.5 to 10.0 and temperature not exceeding 50 degrees C. The enzyme also demonstrated different substrate specificity from known esterases, showing activities towards a broad range of phenolic esters.

The results showed that CRL1446 and CRL1231 powders were able to resist prolonged storage and GIT conditions, retaining FE activity and preserving their probiotic potential.

Summary: This study analyzed the changes in the human gut microbiota during a fecal microbiota transplantation (FMT) trial using nanopore technology. The results show that the engraftment of donors' microbiota is correlated with the improvement of metabolic health in recipients. Furthermore, the study revealed that assessing nucleotide frequencies can successfully explore the engraftment of donor bacterial strains. The findings indicate that factors such as baseline gut microbiota configuration, sex, and age of donors should be considered for successful FMT in humans.

Summary: In this study, three FAE+ L. johnsonii strains were isolated and compared as silage inoculants. The results showed that L. johnsonii ETC187 had better inoculant and probiotic abilities. Although L. johnsonii ETC187 significantly reduced ADF, L. johnsonii ETC150 and CRL1446 not only induced similar ADF reduction, but also reduced dry matter loss.

Summary: This study aimed to design functional fermented goat milk with probiotic potential for metabolic diseases. The researchers characterized indigenous lactobacilli from goat dairy products that target improving the inflammatory, lipid, and glycemic profile. Fermented goat milk incorporating these lactobacilli showed positive effects on inhibiting alpha-glucosidase, bile salts hydrolase activity, cholesterol assimilation, and reducing triglyceride percentage. The oral administration of these lactobacilli to obese mice resulted in significant weight loss and improved hyperglycemia and hyperlipemia.
