Insertion of an Alu element in a lncRNA leads to primate-specific modulation of alternative splicing

Noncoding RNAs, mobile elements, and alternative splicing are all critical for the regulation of gene expression. Here we show that a conserved noncoding RNA acquires a new function due to the insertion of a mobile element. We identified a noncoding RNA, termed 5S-OT, which is transcribed from 5S rDNA loci in eukaryotes including fission yeast and mammals. 5S-OT plays a cis role in regulating the transcription of 5S rRNA in mice and humans. In the anthropoidea suborder of primates, an antisense Alu element has been inserted at the 5S-OT locus. We found that in human cells, 5S-OT regulates alternative splicing of multiple genes in trans via Alu/anti-Alu pairing with target genes and by interacting with the splicing factor U2AF65. This trans effect of 5S-OT in splicing might be exploited in biotechnological applications.

Reciprocal modulation of long noncoding RNA EMS and p53 regulates tumorigenesis

Summary: This study reveals a mutual repression between p53 and lncRNA E2F1 messenger RNA stabilizing factor (EMS) in tumor suppression. EMS inhibits p53 expression by disrupting the CPEB2-p53 mRNA interaction, leading to a suppression of p53 translation. EMS plays a critical role in promoting tumorigenesis through negative regulation of p53.

Circular RNAs in physiology and non-immunological diseases

Summary: Circular RNAs (circRNAs) are covalently closed single-stranded RNAs with unique features in their biogenesis, degradation, and transport. They have diverse molecular functions, including sponging miRNAs, regulating transcription, modulating RNA-binding proteins, and even encoding proteins. Dysregulation of circRNAs is associated with various human disorders. Further research can help resolve debates and potentially lead to biomedical applications of circRNAs.

Emerging functions of mitochondria-encoded noncoding RNAs

Summary: Mitochondria, organelles with circular genomes, play critical roles in energy production and homeostasis maintenance in eukaryotic cells. Recent studies have identified various mitochondria-encoded noncoding RNAs (mt-ncRNAs), including novel types such as mecciRNAs and mt-dsRNAs. This review discusses the expression patterns, biogenesis, metabolism, regulatory roles, and functional mechanisms of mt-ncRNAs, highlighting their importance in cellular physiology and potential involvement in human diseases. Additionally, advancements in technologies and methodologies, as well as future perspectives and challenges in studying these noncoding RNAs and their biomedical applications, are also addressed.
