Detection of aberrant methylated SEPT9 and NTRK3 genes in sporadic colorectal cancer patients as a potential diagnostic biomarker

Colorectal cancer (CRC) is one of the most common malignancies, and the third leading cause of cancer mortality worldwide. Timely detection of CRC in patients with earlier stages provides the highest rate of survival. Epigenetic alterations are important in the occurrence and progression of CRC, and represent the primary modifications of cancer cells. Therefore, detection of these alterations in CRC cases are thought to hold great promise as diagnostic biomarkers. It has been shown that the SEPT9 and NTRK3 genes are aberrantly methylated and their detection can be used as biomarkers for early diagnosis of CRC. The present study analyzed promoter methylation status of these genes in CRC patients. Genomic DNA was extracted from 45 CRC and paired adjacent healthy tissues and undergone bisulfite conversion, and the methylation status of NTRK3 and SEPT9 were defined using the MS-HRM assay. Our results showed that there are statistically significant differences in methylation status of NTRK3 and specially SEPT9 between CRC and adjacent normal tissues (P<0.001). High sensitivity and specificity for a specific location in SEPT9 gene promoter as a diagnostic biomarker was observed. SEPT9 promoter hypermethylation may serve as a promising biomarker for the detection of CRC development. However, to validate the biomarker potential of NTRK3 there is a requirement for further investigation.

Computational tools were used to validate the biological activities of the vaccine, predict the binding of the protein candidate with Toll-like receptor 4 and HLA-A*11:01, and molecular dynamics simulation was employed to evaluate the stability of the designed fusion protein with the receptors.

PAX2 promoter methylation and AIB1 overexpression promote tamoxifen resistance in breast carcinoma patients

Summary: This study found that aberrant promoter methylation of PAX2 and overexpression of AIB1 are associated with tamoxifen response in breast carcinoma patients, which may serve as predictive or prognostic biomarkers. Further research is needed to explore the potential of using PAX2 and AIB1 expression and their methylation-mediated regulation as a new target therapy for better disease management.

Multi-targeting of K-Ras domains and mutations by peptide and small molecule inhibitors

Summary: K-Ras activating mutations are associated with tumor progression and metastasis. We aimed to target this mutated protein using small peptides and small molecules. Through computational methods, we selected peptides and small molecules that showed strong binding affinity to mutated and oncogenic K-Ras. These compounds hindered the growth of pancreatic tumor cells and reduced the expression of downstream genes. Our combinatorial approach introduces new potential candidates for blocking oncogenic K-Ras in various cancers. Further in vitro and in vivo analysis is needed to validate the efficacy and safety of these compounds.

The results showed elevated levels of VEGF-C and PTGS2, which correlated with WBC, Platelet, and Hemoglobin levels. Overexpression of VEGF-C and PTGS2 was associated with poorer survival in patients.

Summary: This review investigates the impact of promoter hypermethylation on key tumor suppressor genes involved in cell growth and apoptosis in thyroid cancer. The aberrant epigenetic methylation of certain tumor suppressor genes may have potential clinical implications by activating kinase pathways, while other genes may suppress malignant cell proliferation by inhibiting the cell cycle and inducing apoptosis.
