Leptin changes differentiation fate and induces senescence in chondrogenic progenitor cells

Body weight is a component of the mechanical theory of OA (osteoarthritis) pathogenesis. Obesity was also found to be a risk factor for digital OA involving non-weight-bearing joints, which suggested that metabolism influences the occurrence and progression of OA. The metabolic origin of OA has been partially attributed to the involvement of adipokines, such as leptin, the levels of which are significantly and positively correlated with cartilage degeneration in OA patients. However, the mechanisms by which leptin-induced cartilage degeneration occurs are poorly understood. The discovery of chondrogenic progenitor cells (CPCs) opened up new opportunities for investigation. Investigating the effects of leptin on differentiation and proliferation in CPCs would increase our understanding of the roles played by leptin in the aetiology and development of OA. Here, CPCs were harvested using single-cell sorting from rat cartilage tissues to obtain mesenchymal stem-like cells, which possess clonogenicity, proliferation and stemness. High doses of leptin decreased the ability of the CPCs to migrate, inhibited their chondrogenic potential and increased their osteogenic potential, suggesting that leptin changes differentiation fates in CPCs. High doses of leptin induced cell cycle arrest and senescence in CPCs by activating the p53/p21 pathway and inhibiting the Sirt1 pathway. Inhibiting the Sirt1 pathway accelerated cartilage senescence in knockout (KO) mice. Activating the leptin pathway induced higher Ob-Rb expression and was significantly correlated with cartilage degeneration (lower levels of Coll-2) and tissue senescence (higher levels of p53/p21 and lower levels of Sirt1) in OA patients, suggesting that leptin-induced CPCs senescence contributes to the development of OA. Taken together, our results reveal new links between obesity and cartilage damage that are induced by leptin-mediated effects on cell behaviour and senescence.

A combined finite element and deep learning network for structural dynamic response estimation on concrete gravity dam subjected to blast loads

Summary: This paper develops advanced deep learning approaches for structural dynamic response prediction and dam health diagnosis. Improved long short-term memory (LSTM) networks are proposed for data-driven analysis of structural dynamic responses. The LSTM-based models are proven to have the potential for quick estimation of structural responses under blast loads. Damage indicators extracted from the predicted velocity histories establish their relationship with the numerical simulation of dam damage. This study provides a deep-learning based framework for quick assessment of dam explosions.

A science mapping-based review of work-related musculoskeletal disorders among construction workers

Group Consensus of Second-Order Sample MASs via Distributed Event-Triggered Mechanism

Summary: The group consensus of second-order sample multi-agent systems is investigated using a distributed event-triggered mechanism. A consensus protocol is proposed, which is updated only when the event-triggered condition is met and the update only depends on the data collected at the moment of triggering. The sufficient condition for group consensus is obtained, and a differential equation is constructed to avoid the Zeno phenomenon and obtain a minimum positive and lower bound for any two trigger time intervals. The effectiveness is verified through a simulation example.

Experimental Study on Bond Performance of NC-UHPC Interfaces with Different Roughness and Substrate Strength

Summary: This paper investigates the bond failure performance between precast normal concrete (NC) and cast-in situ ultra-high performance concrete (UHPC), emphasizing the influence of interfacial roughness. The results demonstrate that certain interfacial roughness is essential to gain a reliable bond connection between NC and UHPC. The higher strength of the base concrete is beneficial to the bond performance, but the influence on the shear interfacial performance could be offset by that of the interface morphology in the case with high interfacial roughness.

Summary: This study aimed to investigate the experiences, challenges, concerns, and coping strategies of older women with chronic low back pain (CLBP) in Hong Kong. The findings revealed that CLBP has negative impacts on older women's daily life, both physically and psychologically. They adopted various pain management strategies, with some influenced by the Chinese culture. Family support, as well as social activities and support from elderly community centers, were found to be important factors for their management of CLBP.

Strain-Hardening, impact protective and Self-Healing supramolecular polyurethane nanocomposites enabled by quadruple H-Bonding, disulfide bonds and nanoparticles

Summary: Developing protective materials with intelligent and stimuli-responsive properties is essential for safety purposes. However, combining contradictory properties into a single polymer is challenging. In this study, a strain-hardening supramolecular polyurethane nanocomposite (SPN) with high mechanical strength and unique impact protection properties is synthesized by incorporating quadruple hydrogen bonds (H-bonds), disulfide bonds, and modified silica nanoparticles into polyurethane elastomer. This SPN material exhibits high energy absorption efficiency and impact protection properties through the dynamic break and re-form characteristic of quadruple H-bonds and the stress dissipation behavior of nanoparticles.

Recorded low-reflection in gradient MXene-decorated melamine microwave shielding foam integrated with piezoresistive sensing and energy absorption properties

Summary: High-performance electromagnetic interference (EMI) shielding materials with low reflection are urgently needed to address secondary electromagnetic pollution. This study proposes ecoflex-encased gradient MXene-decorated melamine foams (eGMMFs) to achieve both ultra-low reflection and efficient shielding. By designing a gradual increased gradient MXene conductive network and a reflector of reduced graphene oxide, the eGMMF integrates multiple layers for improved impedance matching, loss, and reflection, resulting in an average shielding efficiency of 78.8 dB and a reflection coefficient (R) of 0.022 in the X-band. A record-low R value of 3.84 x 10(-5) at 11.52 GHz is achieved, surpassing previously reported MXene-based shields. Furthermore, the developed eGMMF exhibits remarkable piezoresistive sensing and anti-impacting performance. This work provides new insights and guidance for the development of multifunctional shielding and absorbing materials.

A fast end-to-end method for automatic interior progress evaluation using panoramic images

Summary: This paper introduces a method based on 360 degrees panoramic images and deep learning for fast end-to-end interior progress evaluation. The proposed method takes only one or two 360 degrees panoramic images as input, estimates key corners, generates and registers room layouts, and semantically segments sparse point cloud. The experimental results show that the proposed method achieves comparable performance against those on public data sets.

Exploring the adoption of blockchain in modular integrated construction projects: A game theory-based analysis

Summary: Efficient information sharing is crucial for the success of Modular integrated Construction (MiC) projects. Blockchain provides powerful solutions to address information barriers and foster trust among MiC stakeholders. However, the construction industry lacks empirical evidence and is cautious about adopting this disruptive technology. This study uses game theory to examine stakeholders' decision-making processes and identifies key factors influencing their decisions, such as diffusion rate value thresholds, benefits of blockchain adoption, government subsidies, and relevant costs. The study also suggests pilot projects and subsidy or incentive policies to enhance stakeholders' perceived values and promote blockchain adoption in MiC projects.

Cooperation mode for concrete formwork reuse among construction sites

Summary: Concrete formwork reuse can save costs and reduce resource consumption, and it has recently gained more attention. Cooperation among construction sites in a specific district can facilitate the reuse of idle or underutilized formwork. However, the cooperation method and profit allocation are rarely explored, and the assumption of no contract violations in cooperation is not aligned with reality. This study proposes an applicable cooperation mode that includes optimal alliance establishment, profit allocation based on contracts, and compensation for contract violations.

Exploring the impact of policy interventions on project performance through a PSM-DID approach: evidence from the Hong Kong construction industry
