Summary: In the field of clinical immunology, the outbreak of COVID-19 has driven studies on the role of inflammation in cardiovascular diseases. Advanced molecular genetic methods have been used to explore the diagnostic significance of these diseases and to identify immunological interactions at organ and system levels. Additionally, research has shown the influence of emotional stress on viral and cardiovascular disorders through brain-immune system interactions. However, there are challenges that need to be addressed before these findings can be fully applied in the clinical arena.

Summary: Although the role of adaptive SARS-CoV-2 specific immunity in post-acute sequelae of COVID-19 (PASC) is not well explored, this study found that PASC patients have a stronger CD8+ T cell response and comparable neutralizing capacity compared to controls. The persistent inflammatory response triggered by low avidity SARS-CoV-2 reactive CD8+ T cells may be responsible for the observed sequelae in PASC patients. Further studies are needed for a better understanding of the underlying immunopathogenesis.

Summary: Ischemic cardiomyopathy, driven by loss of cardiomyocytes and inadequate proliferative response, is a major global health problem. Functional high-throughput screening revealed that overexpression of certain miRNAs, especially miR-515-3p and miR-519e-3p from the C19MC-cluster, enhanced proliferative activity in hiPSC-CM and altered signaling pathways relevant for cardiomyocyte proliferation.

Observational Study of Repeat Immunoadsorption (RIA) in Post-COVID ME/CFS Patients with Elevated ß2-Adrenergic Receptor Autoantibodies-An Interim Report

Summary: There is increasing evidence suggesting an autoimmune cause for post-infectious Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS), with SARS-CoV-2 being the main trigger. A small proof-of-concept study on IgG depletion using immunoadsorption (IA) showed efficacy in most patients with post-infectious ME/CFS. An ongoing observational study aims to evaluate the efficacy of IA in post-COVID-19 ME/CFS patients.

Exploration of the Noncoding Genome for Human-Specific Therapeutic Targets-Recent Insights at Molecular and Cellular Level

Summary: While most of the human genome does not encode proteins, it is still transcriptionally active and produces a variety of noncoding RNAs with complex functions. Only a small fraction of these transcripts have been assigned specific functions. Research on the noncoding genome has revealed the complexity of the human genome and raised many unanswered questions. Recent studies suggest that noncoding RNAs could serve as novel therapeutic targets for human diseases and may be linked to brain evolution and the interaction between the brain and immune system.

A novel Troponin I mutation associated with severe restrictive cardiomyopathy-a case report of a 27-year-old woman with fatigue

Summary: Restrictive cardiomyopathy is a rare and heterogeneous disease, and this case report presents a familial form of restrictive cardiomyopathy associated with a novel TNNI3 gene mutation. The patient had an oligosymptomatic early presentation and the diagnosis was complicated. Longitudinal follow-up and genetic testing played important roles in understanding the disease. Broad genetic testing should be considered early in the diagnostic workflow.
