Genetic variability of Rickettsia spp. in Ixodes persulcatus ticks from continental and island areas of the Russian Far East

Rickettsia spp. are intracellular Gram-negative bacteria transmitted by arthropods. Two potentially pathogenic rickettsiae, Candidatus Rickettsia tarasevichiae and Rickettsia helvetica, have been found in unfed adult Ixodes persulcatus ticks. The aim of this study was to assess the prevalence and genetic variability of Rickettsia spp. in I. persulcatus ticks collected from different locations in the Russian Far East. In total, 604 adult I. persulcatus ticks collected from four sites in the Khabarovsk Territory (continental area) and one site in Sakhalin Island were examined for the presence of Rickettsia spp. by real-time PCR. Nested PCR with species-specific primers and sequencing were used for genotyping of revealed rickettsiae. The overall prevalence of Rickettsia spp. in ticks collected in different sites varied from 67.9 to 90.7%. However, the proportion of different Rickettsia species observed in ticks from Sakhalin Island significantly differed from that in ticks from the Khabarovsk Territory. In Sakhalin Island, R. helvetica prevailed in examined ticks, while Candidatus R. tarasevichiae was predominant in the Khabarovsk Territory. For gltA and ompB gene fragments, the sequences obtained for Candidatus R. tarasevichiae from all studied sites were identical to each other and to the known sequences of this species. According to sequence analysis of gltA, ompB and sca4 genes, R. helvetica isolates from Sakhalin Island and the Khabarovsk Territory were identical to each other, but they differed from R. helvetica from other regions and from those found in other tick species. For the first time, DNA of pathogenic Rickettsia heilongjiangensis was detected in I. persulcatus ticks in two sites from the Khabarovsk Territory. The gltA, ompA and ompB gene sequences of R. heilongjiangensis were identical to or had solitary mismatches with the corresponding sequences of R. heilongjiangensis found in other tick species. The EC151 genome is 60,753 bp in size, containing 58 putative genes, with limited similarity to other phage genomes in the NCBI GenBank database. Comparative proteomic phylogenetic analysis places EC151 in a distinct branch within a group of sequences sharing clades with Seuratvirus, Nonagvirus, and Vidquintavirus genera.

Occurrence and clinical manifestations of tick-borne rickettsioses in Western Siberia: First Russian cases of Rickettsia aeschlimannii and Rickettsia slovaca infections

Summary: This study aimed to determine the etiologic agents of tick-borne rickettsioses in Western Siberia during three epidemiological seasons. The findings showed that Rickettsia raoultii is a common agent of tick-borne rickettsioses in Novosibirsk Province, and DNA from Rickettsia aeschlimannii and Rickettsia slovaca was found in clinical samples of patients in the Russian Federation for the first time.

Antibiotic Resistance and Pathogenomics of Staphylococci Circulating in Novosibirsk, Russia

Summary: A study in Novosibirsk, Siberian Russia, characterized 394 strains of staphylococci from humans and pets in terms of antibiotic resistance and genes, revealing some sensitive isolates but prevalent methicillin-resistant and multidrug-resistant strains across all major species. Specific genes associated with resistance were identified, indicating a complex pattern of multidrug resistance, with additional antibiotic resistance genes found in MDR strains. Additionally, biofilm-producing abilities and genes related to virulence and pathogenicity were more prominent in MDR S. epidermidis compared to S. haemolyticus.

Summary: A new candidate species of the spotted fever group, Candidatus Rickettsia thierseensis, was found in Austria and its gene sequences were highly similar to Candidatus Rickettsia uralica identified in Russia. Summary: Two novel P. protegens bacteriophages, PseuP_222 and Pseu_224, were isolated along with their host P. protegens CEMTC 4060 from the Inya river in Siberia. These phages have a siphovirus morphology and belong to lambdoid phages. Comparative genome analysis revealed their low similarity to other lambdoid phages and indicated that they belong to a genetically diverse group of phages of environmental Pseudomonas spp. The substantial divergences in their genomes and proteomes suggest that these phages have evolved independently and have recently started using the same host.

Novel Genetic Lineages of Rickettsia helvetica Associated with Ixodes apronophorus and Ixodes trianguliceps Ticks

Summary: The prevalence and genetic diversity of Rickettsia spp. were investigated in Ixodes apronophorus, Ixodes persulcatus, and Ixodes trianguliceps ticks from the same habitats in Western Siberia. Rickettsia helvetica was found in I. apronophorus, while Candidatus Rickettsia tarasevichiae dominated in I. persulcatus and I. trianguliceps were infected with Candidatus Rickettsia uralica, R. helvetica, and Ca. R. tarasevichiae. Phylogenetic analysis revealed the high genetic variability of R. helvetica.

Antibodies Capable of Enhancing SARS-CoV-2 Infection Can Circulate in Patients with Severe COVID-19

Summary: This study identified the first monoclonal antibody (mAb) that can cause antibody-dependent enhancement (ADE) in a SARS-CoV-2 in vivo model. The mAb RS2 showed high affinity for SARS-CoV-2 and demonstrated neutralizing ability in vitro. It was also found that severe COVID-19 patients had antibodies competing with mAb RS2.

Genomes of a Novel Group of Phages That Use Alternative Genetic Code Found in Human Gut Viromes

Summary: In this study, a unique phage genome was discovered during the analysis of the human gut virome. Comparative analysis identified two similar phage genomes, indicating the presence of a new species within the proposed Phassvirus genus. Furthermore, a group of divergent phage genomes including the proposed Phassvirus genus was identified in metagenome data. These findings highlight the importance of using different translation tables when depositing phage genomes in GenBank.

Isolation, Characterization and Genomic Analysis of a Novel Jumbo Phage, AerS_266, That Infects Aeromonas salmonicida

Summary: Aeromonas salmonicida is a causative agent of septicemia in fish, leading to significant economic losses in the aquaculture industry. The emergence of antibiotic resistance requires the development of alternative treatment methods, such as phage therapy. In this study, a novel A. salmonicida jumbo phage, AerS_266, was isolated and characterized. It specifically infects mesophilic A. salmonicida strains and has a slow lytic life cycle. The phage's genome contains 253 putative genes, including genes encoding two multisubunit RNA polymerases, chimallin and PhuZ, making it a phiKZ-like phage. This is the first phiKZ-like phage found to infect A. salmonicida, although similar phages have been described for other Aeromonas species.

Babesia negevi infection in dogs and response to treatment

Summary: This study describes the presentation, clinical signs, parasitemia levels, laboratory findings and treatment of dogs infected with B. negevi. Treatment with imidocarb dipropionate was helpful for recovery from clinical disease but did not facilitate parasite elimination, and it is therefore recommended to treat canine B. negevi infection with the combination of atovaquone and azithromycin.

Tick-borne pathogens in camels: A systematic review and meta-analysis of the prevalence in dromedaries

More research is needed to understand the role of camels in maintaining and transmitting these pathogens.

Original Identifying breeding hosts of Ixodes ricinus ticks using stable isotope analysis of their larvae - Proof of concept
