Vitreous Cryopreservation of Human Umbilical Vein Endothelial Cells with Low Concentration of Cryoprotective Agents for Vascular Tissue Engineering

Cryopreservation of human umbilical vein endothelial cells (HUVECs) is important to tissue engineering applications and the study of the role of endothelial cells in cardiovascular and cerebrovascular diseases. The traditional methods for cryopreservation by vitrification (cooling samples to a cryogenic temperature without apparent freezing) using high concentration of cryoprotective agents (CPAs) and slow freezing are suboptimal due to the severe toxicity of high concentration of CPAs and ice formation-induced cryoinjuries, respectively. In this study, we developed a method to cryopreserve HUVECs by vitrification with low concentration of CPAs. This is achieved by optimizing the CPAs and using highly thermally conductive quartz capillary (QC) to contain samples for vitrification. The latter minimizes the thermal mass to create ultra-fast cooling/warming rates. Our data demonstrate that HUVECs can be vitrified in the QC using 1.4mol/L ethylene glycol and 1.1mol/L dimethyl sulfoxide with more than 90% viability. Moreover, this method significantly improves the attachment efficiency of the cryopreserved HUVECs. The attached cells post-cryopreservation proliferate similarly to fresh cells. Therefore, this study may provide an effective vitrification technique to bank HUVECs for vascular tissue engineering and other applications.

In this study, Polycaprolactone (PCL) nanofibers with nano-tantalum oxide (Ta2O5) were successfully fabricated through electrospinning. The chemical reaction between hydrophobic scaffolds and acid oxides in the mineralization process has a significant impact on the biological effects of the scaffolds.

Hydrogel Microencapsulation Enhances Cryopreservation of Red Blood Cells with Trehalose

Summary: This study explored a microencapsulation method for cryopreservation of red blood cells (RBCs). The results showed that the recovery of RBCs after microencapsulation was significantly improved, and the washing process was simplified. The microencapsulation method provides a promising, referenceable, and more practical strategy for future clinical transfusion medicine.

Automatic Classification of Red Blood Cell Morphology Based on Quantitative Phase Imaging

This study combines quantitative phase imaging (QPI) technology with deep learning to automatically classify RBC morphology. The developed method shows higher accuracy and shorter processing time compared to traditional methods, especially for RBCs with more discocytes.

Mechanical Gradients Enable Highly Stretchable Electronics Based on Nanofiber Substrates

Summary: In this study, a mechanical gradient strategy was developed to fabricate high-performance stretchable electronic devices. By using polyvinyl alcohol glue to fix integrated circuits on stretchable circuits, the strain gradient during the stretching process was achieved, ensuring the stability and reliability of the devices. The flexible electronic devices fabricated by this method are highly stretchable and comfortable, making them suitable for various applications.

Summary: Ovarian follicles develop in a regulated mechanical microenvironment, and disruptions to this microenvironment can cause infertility. The viscoelastic properties of ovarian tissue have not been extensively studied. This study characterizes the elastic and viscoelastic properties of ovarian tissue from both reproductive age groups of domestic cats and identifies possible connections between mechanical and compositional heterogeneities. These findings may have important implications for biomimetic follicle culture.

Static magnetic field assisted thawing improves cryopreservation of mouse whole ovaries

Summary: This study proposes a new method to improve the antioxidant capacity of whole ovaries during cryopreservation, static magnetic field assisted thawing. The results show that the antioxidant capacity of ovarian tissue was significantly improved by static magnetic field treatment. In addition, successful vascular regeneration and follicular development were achieved through ovarian tissue allograft transplantation.
