Summary: Aromatase inhibitors (AIs) are essential in the management of hormone receptor-positive (HR+) breast cancer, but their use is limited by the musculoskeletal adverse effects known as aromatase inhibitor-associated musculoskeletal syndrome (AIMSS), which includes bone loss and arthralgias. The pathophysiology of AIMSS is not fully understood, but is proposed to be related to estrogen deprivation in the musculoskeletal and nervous systems.

Summary: The study highlights the association between mitochondrial alterations and AI resistance in breast cancer patients, indicating that changes in mitochondrial function could impact patient prognosis. Induction of drug resistance in breast cancer cells through mitochondrial stress adaptation, possibly due to the activation of the AREG-ER alpha loop signaling, suggest potential treatment targets for overcoming AI resistance.

Summary: The study found that aromatase expression in Leydig cells was increased in men with nonobstructive azoospermia (NOA) and significantly associated with intratesticular testosterone (ITT) and estradiol (ITE2) levels, as well as sperm retrieval success. Treatment with anastrozole increased the ITT/ITE2 ratio and decreased aromatase expression, potentially offering a therapeutic window for selected patients with azoospermia.

Summary: Aromatase inhibitor-induced arthralgia (AIA) is characterized by significant musculoskeletal symptoms such as joint pain, myalgia, and joint stiffness. A prospective study conducted on postmenopausal women with early-stage hormone receptor positive (HR+) breast cancer found that changes in tendon and median nerve velocities were associated with pain and stiffness levels. Higher velocities were linked to greater pain, while slower velocities were associated with higher stiffness scores. Increased velocity in the abductor pollicis longus tendon was found to be related to worsened pain and stiffness measures.

Summary: Aromatase inhibitor-induced arthralgia (AIA) is a common problem in breast cancer patients and affects their adherence to treatment. This systematic review and network meta-analysis evaluated the effectiveness of different therapies for AIA. The results showed that acupuncture ranked the highest in improving pain intensity, followed by sham acupuncture, herbal medicine, exercise, duloxetine, vitamin D, omega-3 fatty acids, physical therapy, testosterone, and inactive controls. This review provides valuable insights for evidence-based decision making in the management of AIA in breast cancer survivors.

Summary: This study aimed to assess the relationship between physical activity (PA) and pain, and also examine the prevalence of pain, PA levels, health care seeking behaviors, and impact of pain on daily activities and work. The results showed that individuals who met the PA guidelines had lower odds of reporting pain and engagement in PA decreased with higher pain frequency.

Summary: Population aging, high prevalence of non-communicable diseases, physical inactivity, and rising global temperatures are major public health concerns. Regular physical activity throughout life can play an important role in adapting to rising temperatures, coping with heat-related health risks, and increasing individual and community resilience. This viewpoint calls for more research on the contribution of physical activity to adapting to rising global temperatures and climate change.

Summary: This study aimed to assess the sexual health of breast cancer survivors and explore factors associated with sexual inactivity and reduced sexual functioning. The results showed that half of breast cancer survivors were sexually inactive, with lack of interest being the most common reason. Treatment with aromatase inhibitor and poor body image were associated with sexual inactivity.

Summary: Decision aids with values clarification exercises can help reduce opposition and increase support for genomic sequencing for newborns, as shown by a study on parental beliefs and intentions. Participants who engaged in values clarification exercises showed weaker agreement with statements against sequencing and stronger agreement with reasons supporting sequencing, influencing their intentions to have their newborn sequenced.

Summary: This study examined the effects of a mobile health (mHealth) physical activity intervention on 280 young adult cancer survivors (YACS). The results showed that after 12 months, there were no differences in accelerometer-measured total physical activity time between the intervention and self-help groups, but the intervention group had a greater increase in self-reported total physical activity time. Both groups maintained their physical activity levels from 6 to 12 months, and at 12 months, more intervention participants met the national physical activity guidelines compared to the self-help participants.
