Study Design.A cross-sectional study of 2 groups of patients with scoliosis, and an age-matched control group was conducted. Each of the groups such as patients with adolescent idiopathic scoliosis (AIS) as well as control group were divided additionally into 2 groups: premenarcheal and postmenarcheal girls.Objective.The aim of the study was to determine the levels of 25-OH-vitamin D3, calcium and phosphate, parathyroid hormone (PTH), and calcitonin in serum of pre- and postmenarcheal girls with AIS and corresponding groups of scoliosis-free controls.Summary of Background Data. The primary etiology and pathogenesis of AIS remains unknown. It is assumed that vitamin D deficiency and genetic predisposition, for example, polymorphisms of vitamin D receptor, have a great significance. Vitamin D plays a key role in skeletal development and prevents bone atrophy, affects the absorption of calcium, maintains calcium-phosphate homeostasis, and the bone matrix mineralization. Its deficiency can result in a wide variety of skeletal deformities, low bone mass, and then leads to the disappearance of bone. Defects in trabecular bone structure and/or bone mineralization are the main features of scoliosis. Some studies have reported that Vitamin D deficiency is common among patients with AIS. The mechanism of Vitamin D action on scoliosis development is still unclear.Methods.Determination of serum 25-OH-D3 levels was performed using high-performance liquid chromatography chromatography; concentrations of calcium and phosphate were measured using colorimetric methods, and concentration of PTH and calcitonin was measured using ELISA system.Results.Reduction in the serum levels of 25-OH-D3 and calcitonin in girls with AIS compared with healthy girls was demonstrated.Conclusion.The phosphate-calcium balance and PTH level seem to be normal in patients with AIS. The calcitonin level in girls with AIS is 2-fold lower than in healthy subjects. It is possible that the deficiency of vitamin D can be involved in AIS.Level of Evidence: 4

Oncogenic translation directs spliceosome dynamics revealing an integral role for SF3A3 in breast cancer

Maciej Ciesla, Phuong Cao Thi Ngoc, Eugenia Cordero, Alvaro Sejas Martinez, Mikkel Morsing, Sowndarya Muthukumar, Giulia Beneventi, Magdalena Madej, Roberto Munita, Terese Jonsson, Kristina Lovgren, Anna Ebbesson, Bjorn Nodin, Ingrid Hedenfalk, Karin Jirstrom, Johan Vallon-Christersson, Gabriella Honeth, Johan Staaf, Danny Incarnato, Kristian Pietras, Ana Bosch, Cristian Bellodi

Summary: The study uncovers the central role of the core splice factor SF3A3 in rewiring splicing during malignant transformation, particularly in the context of MYC hyperactivation. SF3A3 levels are modulated through translation in an elF3D-dependent manner, leading to metabolic reprogramming and stem-like properties that fuel MYC tumorigenic potential. Overall, these findings reveal a post-transcriptional interplay between splicing and translation that governs critical facets of MYC-driven oncogenesis.

Summary: High-sucrose diet influences the mRNA levels and enzyme activities of elongases and desaturases in rat adipose tissue, leading to changes in fatty acid composition. Specifically, Elovl5, Elovl6, Fads1, and Scd mRNA levels were higher in HSD rats, while Fads2 mRNA level was lower. Monounsaturated fatty acids were also decreased in the AT of HSD rats compared to the ST group.

Summary: Positive effects of music therapy for people with dementia and their family carers are reported in studies, but more adequately powered evidence-based studies are needed. Recruitment in dementia trials is complex and challenging and needs thorough planning. Future studies should include reporting of recruitment strategies and related aspects to aid in better design and resource estimation.

Summary: This study demonstrates that pseudouridylation of a stem cell-enriched tRF subtype, mini tRFs containing a 5' terminal oligoguanine (mTOG), selectively inhibits aberrant protein synthesis programs, thereby promoting engraftment and differentiation of hematopoietic stem and progenitor cells in MDS patients. The mechanism involves hindering the recruitment of translational co-activator PAIP1 and repressing translation of transcripts sharing pyrimidine-enriched sequences at the 5' UTR, including TOP sequences frequently altered in cancer. Dysregulation of mTOG leads to increased translation of 5' PES mRNA in malignant MDS-HSPCs, which is associated with leukaemic transformation and reduced patient survival.

Self at Risk: Self-Esteem and Quality of Life in Cancer Patients Undergoing Surgical Treatment and Experiencing Bodily Deformities

Ewa Wojtyna, Malgorzata Pasek, Aleksandra Nowakowska, Anna Gozdzialska, Malgorzata Jochymek

Summary: Self-esteem is an important factor affecting the quality of life (QoL) for cancer patients undergoing surgical procedures for bodily deformities. However, little data exists on which aspects of self-esteem are most closely associated with QoL. This article presents two studies aimed at addressing this gap. Study 1 focuses on changes in global self-esteem and QoL in oral cancer patients (n = 35) treated surgically, while Study 2 examines changes in explicit and implicit self-esteem and QoL in women with breast cancer undergoing mastectomy (n = 96). Both studies used the EORTC QLQ-C30 and Rosenberg's SES questionnaires, with Study 2 also including the Implicit Association Test (IAT). Results showed that patients experienced a decline in global QoL and self-esteem after surgery. Study 1 revealed that patients with higher initial self-esteem experienced a greater decrease in self-esteem and QoL compared to those with lower initial self-esteem. In Study 2, women with fragile self-esteem exhibited the largest decreases in various dimensions of QoL and explicit self-esteem. Conversely, women with high explicit and implicit self-esteem had the best QoL after mastectomy. These findings highlight the need for special psycho-oncological care for cancer patients with high, fragile self-esteem.
