Summary: In this study, the researchers found that the E3 ubiquitin ligase LNX2 plays an important role in early embryonic development by regulating Notch signaling through the degradation of NUMB protein. Knockdown of LNX2 resulted in the majority of embryos failing to develop into normal blastocysts and increased expression levels of genes involved in cell lineage specification. These findings suggest that LNX2 may be involved in transcriptionally regulating various genes implicated in early embryonic development and may act via lineage specification of the inner cell mass.

Summary: This study investigated the changes in the transcriptome of pig blastocysts caused by vitrification using the OC and SOPS systems. The results showed that vitrification with the OC system resulted in fewer genes related to apoptosis and activated genes related to cell proliferation compared to the SOPS system. However, further investigation is needed to understand how these transcriptome differences affect the subsequent developmental ability of the embryos after transfer.

The results of this study can provide valuable information for medical practitioners and patients regarding the success rates of frozen-thawed embryo transfer.

Summary: A comprehensive study of proteome dynamics in non-rodent mammalian embryos developed in vivo was conducted. Proteomic changes in bovine embryos at different developmental stages were analyzed, and distinct patterns were observed. Proteins that increased in abundance during development were associated with RNA metabolism, protein translation, and ribosome biogenesis, while proteins that decreased in abundance were involved in Golgi vesicle transport and protein processing in endoplasmic reticulum. The results provide valuable insights into the molecular mechanisms underlying early embryo development.

Summary: This study reveals the specific alterations in gene expression of embryonic pig embryos during vitrification (VIT) by comparing them with in vitro cultured (IVC) embryos and analyzing their transcriptomes. The identified VIT alterations may reflect the transcriptional signature of embryo cryodamage and the healing process overcoming the impact of VIT. Selected validated genes are potential biomarkers to improve vitrification.
