The ICH M7 guideline describes a consistent approach to identify, categorize, and control DNA reactive, mutagenic, impurities in pharmaceutical products to limit the potential carcinogenic risk related to such impurities. This paper outlines a series of principles and procedures to consider when generating (Q)SAR assessments aligned with the ICH M7 guideline to be included in a regulatory submission. In the absence of adequate experimental data, the results from two complementary (Q)SAR methodologies may be combined to support an initial hazard classification. This may be followed by an assessment of additional information that serves as the basis for an expert review to support or refute the predictions. This paper elucidates scenarios where additional expert knowledge may be beneficial, what such an expert review may contain, and how the results and accompanying considerations may be documented. Furthermore, the use of these principles and procedures to yield a consistent and robust (Q)SAR-based argument to support impurity qualification for regulatory purposes is described in this manuscript. Summary: Hepatotoxicity is a common adverse effect observed from exposure to xenobiotics, particularly in pharmaceutical research where it can lead to drug withdrawals, clinical failures, and discontinuation of drug candidates. More sustainable and informative methods for assessing hepatotoxicity are critically needed.

Summary: Neurotoxicology focuses on studying the adverse effects of exposure to chemical, biological, or physical agents on the nervous system, and there is a need for alternative methods to assess these effects. The research emphasizes on using computer-assisted methods to predict neurotoxicity based on chemical structure, which must be integrated with experimental information to ensure transparency and consistency in assessments.

Structure-Kinetics Relationships of Opioids from Metadynamics and Machine Learning Analysis

Paween Mahinthichaichan, Ruibin Liu, Quynh N. Vo, Christopher R. Ellis, Lidiya Stavitskaya, Jana Shen

Summary: In 2021, the nation witnessed a record high of opioid overdose deaths, primarily due to synthetic opioids such as fentanyl. Understanding the residence time of opioids is crucial for evaluating the effectiveness of naloxone, a FDA-approved reversal agent. This study estimated the residence times of various fentanyl and morphine analogs and compared them with recent measurements of opioid kinetic properties and naloxone inhibitory constants. Microscopic simulations provided insights into the binding mechanism and molecular determinants of dissociation kinetics for fentanyl analogs, leading to the development of a machine learning approach for analyzing the kinetic impact of fentanyl's substituents.

Summary: In October 2022, the World Health Organization reevaluated the toxic equivalency factors (TEFs) for chlorinated dioxin-like compounds in a panel convened in Lisbon. This effort utilized an updated database, Bayesian dose response modeling, and meta-analysis to derive Best-Estimate TEFs. Applying these new TEFs may result in lower total toxic equivalents for dioxin-like chemicals.
