Lumican Inhibits SNAIL-Induced Melanoma Cell Migration Specifically by Blocking MMP-14 Activity

Lumican, a small leucine rich proteoglycan, inhibits MMP-14 activity and melanoma cell migration in vitro and in vivo. Snail triggers epithelial-mesenchymal transitions endowing epithelial cells with migratory and invasive properties during tumor progression. The aim of this work was to investigate lumican effects on MMP-14 activity and migration of Snail overexpressing B16F1 (Snail-B16F1) melanoma cells and HT-29 colon adenocarcinoma cells. Lumican inhibits the Snail induced MMP-14 activity in B16F1 but not in HT-29 cells. In Snail-B16F1 cells, lumican inhibits migration, growth, and melanoma primary tumor development. A lumican-based strategy targeting Snail-induced MMP-14 activity might be useful for melanoma treatment.

Summary: The study found elevated expression of NMU and NMUR2 in CRC tissue samples and variable expression in CRC cell lines. It was shown for the first time that NMUR2 activation induces signaling in CRC cells, and NMU increases motility and invasiveness in NMUR2-positive CRC cells, as well as promoting prometastatic integrin receptor subunit expression.

Summary: One of the main challenges in the treatment of non-small-cell lung cancer is the development of resistance to cisplatin-based chemotherapy. This study investigates the potential of isothiocyanates (ITCs) and the ALDH inhibitor disulfiram (DSF) as adjuvants to cisplatin chemotherapy. The results show that certain ITCs and DSF can reduce the cisplatin tolerance of resistant lung cancer cells, suggesting their potential in overcoming drug resistance.

Integrated Bioinformatics Analysis of the Hub Genes Involved in Irinotecan Resistance in Colorectal Cancer

Summary: Different drug combinations, including irinotecan, are important in treating colorectal cancer (CRC). However, chemotherapy often leads to drug resistance. This study compared mRNA expression profiles of CRC cell lines and patient samples to select new candidate genes for irinotecan resistance. The results showed that in vitro models have limitations and cannot fully reflect the complex drug resistance network observed in patients.

Neuromedin U secreted by colorectal cancer cells promotes a tumour-supporting microenvironment

Summary: The study revealed that NMU levels are elevated in human CRC compared to normal tissue, with high NMUR1 expression correlating with shorter overall survival in CRC patients. NMUR1 expression was identified in macrophages, endothelial cells, platelets, and platelet microparticles, with NMU-stimulated macrophages increasing the migratory potential of CRC cells. Additionally, NMU activation induced pro-metastatic phenotypes in macrophages and endothelial cells, changing their secretome.

Investigation of Glypican-4 and-6 by Infrared Spectral Imaging during the Hair Growth Cycle

Summary: The expression of glypicans in different hair follicle (HF) compartments was investigated using infrared spectral imaging (IRSI) for the first time, showing the distribution changes of GPC4 and GPC6 in HFs during different phases of the hair growth cycle. Western blot assays confirmed these findings. IRSI demonstrated its ability to identify the location of proteins, proteoglycans, glycosaminoglycans, and sulfated glycosaminoglycans in HFs in a label-free manner.
