Summary: PiwiL1 is not only over-expressed but also plays a major role in tumor induction and progression in cervical cancer. Loss of PiwiL1 decreases tumor-associated properties in CaSki cells, while overexpression of PiwiL1 confers malignant transformation in normal HaCaT cells. Our study reveals a new link between HPV oncogenes, E6 and E7, and PiwiL1. p53 and E2F1 directly bind and differentially regulate the PiwiL1 promoter in a context-dependent manner. Furthermore, RNA-seq and RIP-seq analyses suggest a strong and direct role for PiwiL1 in promoting metastasis in cervical cancer cells.

Summary: This study aims to investigate the expression and regulatory role of hnRNP K in cervical carcinogenesis. The results showed that hnRNP K was highly expressed in cervical cancer and precancerous lesions, and positively correlated with HPV16 E6. hnRNP K induced cell proliferation, inhibited apoptosis, and caused cell cycle arrest in the S phase, as well as increased HPV16 E6 protein expression.

The results showed that MLL deficiency suppressed Kras-induced lung tumorigenesis by decreasing H3K4me3 levels and suppressing the expression of Rasgrf1 gene. Rasgrf1 was essential for the activation of Kras and its downstream pathways, which are involved in cancer-promoting activities.

Summary: In this study, a novel green-synthesized iron oxide-silver nanocomposite-based delivery system for curcumin was developed to improve its druggability against the HPV-16 positive cervical cancer cell line SiHa. The curcumin-nanocomposite showed enhanced anti-proliferative capability compared to free curcumin, with increased cellular uptake at 24 h treatment. It modulated the PI3K/AKT/mTOR signaling cascade to induce both autophagic and apoptotic death in the target cells, mainly through imbalance in intracellular redox system and resultant augmented ROS accumulation. This study suggests that this bimetallic nanocomposite can increase bioavailability and therapeutic efficacy of curcumin.

Summary: PE can restore the impaired function of cervical TIDCs, thereby eliciting further antitumor CTL responses. The effects of PE on TADCs are mediated through inhibition of SOCS1 and activation of downstream JAK2-STAT1/STAT4/STAT5 pathways. PE may be a potent and effective immunomodulatory drug for antitumor treatment via the blockade of SOCS1 signaling in DCs.
