Additionally, SETD1A interacts with and stabilizes beta-catenin to positively regulate the Wnt/beta-catenin pathway.

Summary: Programmed death ligand 1 (PD-L1) plays a crucial role in the development of hepatocellular carcinoma (HCC) by regulating epithelial-mesenchymal transition (EMT) and cancer stem cell (CSC)-like properties. PD-L1 enhances liver CSC phenotypes and tumorigenicity through the activation of stemness genes and the expansion of the CD133(+) cell population. PD-L1 also promotes cell motility, invasion, and EMT process in HCC cells, while suppression of PD-L1 has the opposite effect. PD-L1 interacts with serum and glucocorticoid kinase 2 (SGK2) to activate the SGK2/beta-catenin signaling pathway, thereby inducing EMT and acquisition of a stem cell phenotype in liver cancer cells.

Summary: In this study, we identified BMI1 as a key regulator of spermatogonial stem cell (SSC) maintenance. BMI1 represses target genes by increasing H2AK119ub and reducing H3K4me3 in SSCs. Additionally, BMI1 inhibition activates Wnt signaling and promotes nuclear translocation of beta-catenin in SSCs. Suppression of Wnt/beta-catenin signaling restores SSC maintenance in BMI1-deficient SSCs. Our findings not only reveal a novel mechanism for BMI1 in SSC maintenance, but also provide a potential strategy for treating male infertility.

By binding to EZH2, SNHG9 down-regulated PTEN levels, promoting the self-renewal and tumor formation of liver CSCs and exacerbating hepatocellular carcinoma progression.

Summary: Recent studies have shown that impaired renewal of type 2 alveolar epithelial cells (ATII) in IPF patients and mice with defective ATII can lead to severe pulmonary fibrosis. Growth hormone receptor (GHR) expression, mainly found in mesenchymal cells, is significantly decreased in IPF lungs, but higher levels of GHR expression are associated with better lung function in IPF patients. The findings suggest that a previously unidentified mesenchymal paracrine signaling via GHR can support ATII progenitor cell renewal and limit the severity of lung fibrosis.

Summary: This study identified a deficiency of the zinc transporter SLC39A8 (ZIP8) in Type 2 alveolar epithelial cells (AEC2s) from both idiopathic pulmonary fibrosis (IPF) lungs and lungs of old mice, which is associated with impaired AEC2 renewal capacity and enhanced lung fibrosis. The regulation of AEC2 progenitor function by ZIP8 is dependent on SIRT1. Replenishment with exogenous zinc and activation of SIRT1 promote self-renewal and differentiation of AEC2s, and therapeutic strategies targeting zinc metabolism and SIRT1 signaling could potentially improve AEC2 progenitor function and mitigate fibrogenesis.
