Summary: Cholangiocarcinoma (CCA) is an aggressive type of liver cancer that often metastasizes to the lungs and lymph nodes, resulting in poor patient outcomes. However, the mechanisms behind CCA metastasis are not well understood. In order to study the interactions between CCA tumor cells and the extracellular matrix (ECM) of metastatic sites, researchers created an in vitro model using patient-derived CCA organoids (CCAOs) combined with decellularized human lung and lymph node tissues. This model allowed for the investigation of how the ECM influences metastatic processes and provides new insights into cancer metastasis.

Summary: This study used a spatial characterization method to evaluate tumor-immune interactions and found that the cellular composition of tumor and immune cell subsets in the tumor core was not associated with recurrence, but the spatial patterns were significantly different. It was also found that a cluster enriched with tumor and dendritic cells was associated with no recurrence. Further analysis found an enrichment of CTL-dendritic cell interactions in patients with no recurrence and CTL-macrophage interactions in patients with recurrence.

Summary: This study explored the anti-cancer effects of dandelion seed extract in esophageal squamous cell carcinoma (ESCC) and its impact on the sensitivity of cisplatin treatment. The results showed that dandelion seed extract selectively inhibited cell growth, proliferation, migration, invasion, and angiogenesis in ESCC cells. It also induced cell apoptosis and inhibited epithelial-mesenchymal transition. Furthermore, dandelion seed extract enhanced the sensitivity of ESCC cells to cisplatin by promoting DNA damage and inhibiting the phosphorylation of STAT3.

Summary: Innate lymphoid cells (ILCs), including NK cells and ILC3, play important roles in tissue immune homeostasis and tumor surveillance. In human lymph nodes, NK cells and ILC3s are the prominent subpopulations. However, in tumor-draining lymph nodes and breast cancer tumors, NK cells are more abundant while proportions of ILC3 subsets are low. The local tumor microenvironment inhibits NK cell functions through downregulation of a specific receptor, but cytokine stimulation can restore their functionality.

Summary: RNF149 is upregulated in ESCC and its high expression is associated with poor prognosis. Overexpression of RNF149 confers CDDP resistance to ESCC, but inhibition of RNF149 reverses this phenomenon. Mechanistically, RNF149 interacts with PHLPP2 and induces its degradation, activating the PI3K/AKT signaling pathway. In addition, amplification of RNF149 locus is found in various human cancers. Targeting the RNF149/PHLPP2/PI3K/Akt axis may be a promising prognostic factor and therapeutic target for malignant tumors.
