Toward a Novel Multilocus Phylogenetic Taxonomy for the Dermatophytes

Type and reference strains of members of the onygenalean family Arthrodermataceae have been sequenced for rDNA ITS and partial LSU, the ribosomal 60S protein, and fragments of b-tubulin and translation elongation factor 3. The resulting phylogenetic trees showed a large degree of correspondence, and topologies matched those of earlier published phylogenies demonstrating that the phylogenetic representation of dermatophytes and dermatophyte-like fungi has reached an acceptable level of stability. All trees showed Trichophyton to be polyphyletic. In the present paper, Trichophyton is restricted to mainly the derived clade, resulting in classification of nearly all soudanense, and T. verrucosum. In the newly proposed taxonomy, Trichophyton contains 16 species, Epidermophyton one species, Nannizzia 9 species, Microsporum 3 species, Lophophyton 1 species, Arthroderma 21 species and Ctenomyces 1 species, but more detailed studies remain needed to establish species borderlines. Each species now has a single valid name. Two new genera are introduced: Guarromyces and Paraphyton. The number of genera has increased, but species that are relevant to routine diagnostics now belong to smaller groups, which enhances their identification.

Summary: This study evaluated the in vitro antifungal activity of common and new antifungals against a set of environmental and clinical geophilic dermatophyte isolates, finding that luliconazole, terbinafine, and efinaconazole exhibited the highest efficacy. Further surveillance studies are recommended to confirm the implications of such in vitro data for the clinical recovery rate of dermatophytosis with geophilic species following antifungal therapy.

Summary: This study used a culture-independent niche modeling approach to investigate the ecological adaptations and evolution of Acidobacteria in grassland soils in Germany. The results revealed a pronounced ecological diversification among acidobacterial sister clades and showed that the high diversity of soil acidobacterial communities is largely sustained by differential habitat adaptation. The niche modeling approach correctly predicted the physiological properties of cultivated species of Acidobacteria and provided novel information on ecological adaptations that cannot be inferred from standard taxonomic descriptions.

Sexually acquired pubogenital dermatophytosis induced by Trichophyton quinckeanum

Summary: Tinea corporis is a common superficial fungal infection, whereas tinea of the anogenital region is rare and mostly found in tropical countries. A distinctive variant of pubogenital tinea (PT) characterized by deep tissue infiltration and systemic symptoms has been reported, with transmission via sexual contacts hypothesized. In this case report, PT caused by T. quinckeanum experienced a strong inflammatory reaction following successful treatment with itraconazole.

Post-translational changes in Phialophora verrucosa via lysine lactylation during prolonged presence in a patient with a CARD9-related immune disorder

Summary: CARD9-related inherited immune disorders are associated with chronic disseminated fungal infections, particularly black yeast-like fungi in patients. The genus Phialophora is commonly found in these patients. Genomic analysis revealed accumulation of 6945 SNPs in the derived isolate, with enrichment of proteins involved in melanin biosynthesis. Lactylated proteins play a role in regulating metabolism and pathogenicity in black fungi. Large genomic rearrangements were observed, but not linked to adaptation or clinical properties; in vitro antifungal susceptibility remained largely unchanged.

Proposal to include the categories kingdom and domain in the International Code of Nomenclature of Prokaryotes

Summary: Observations show that adding a category should be done before informal names at that rank are widely used, based on the introduction of the phylum category into the International Code of Nomenclature of Prokaryotes (ICNP). An extrapolation from the number of validly published names under the ICNP at different categories suggests that adding kingdom and domain as separate principal ranks would be beneficial. By examining taxonomic activity and associated nomenclatural problems above the phylum level, it is confirmed that introducing these ranks would help stabilize prokaryotic nomenclature. Proposed amendments to the ICNP include the inclusion of kingdom and domain as additional categories.

Fast and Accurate Identification of Candida auris by High Resolution Mass Spectrometry

A liquid chromatography-high resolution Orbitrap mass spectrometry method has been successfully used for rapid and accurate identification of C. auris strains, providing the possibility for epidemiological surveillance and differentiation between nosocomial transmission and repeated introduction to a hospital.
