ROS-dependent HMGB1 secretion upregulates IL-8 in upper airway epithelial cells under hypoxic condition

High-mobility group box 1 (HMGB1) mediates various functions according to the location. We tried to investigate the role of HMGB1 in upper airway under hypoxic conditions. We cultured primary normal human nasal epithelium (NHNE) cells under hypoxic conditions and evaluated the movement of HMGB1 by western blotting, immunofluorescence, and enzyme-linked immunosorbent assay (ELISA). Reactive oxygen species (ROS) level was evaluated to estimate the translocation mechanism of HMGB1. The role of secreted HMGB1 was evaluated by ELISA assay. Furthermore, we collected human nasal mucosa samples and nasal lavage fluids from patients conditioned under hypoxic and non-hypoxic environment, and compared the expression of HMGB1 in human nasal mucosa samples by immunohistochemistry and the levels of HMGB1 in lavage fluids using ELISA assay. Hypoxia induced translocation of HMGB1 into the extracellular area and it was dependent on ROS produced by dual oxidase 2. Secreted HMGB1 was involved in the upregulation of interleukin (IL)-8. In human samples, HMGB1 was translocated from nucleus to the cytoplasm in hypoxic-conditioned nasal mucosa. HMGB1 was increased in nasal lavage samples of chronic rhinosinusitis patients, whose sinus mucosa was supposed to be hypoxic as compared with controls. We suggest that HMGB1 is secreted in hypoxic condition via ROS-dependent mechanism and secreted HMGB1 participates in IL-8 upregulation mediating inflammatory response.

Serum high-mobility group box 1 protein level correlates with the lowest SaO2 in patients with sleep apnea: a preliminary study

Summary: This study aimed to evaluate the possibility of using the serum high-mobility group box 1 protein level as a biologic marker in obstructive sleep apnea patients. The results showed a significant correlation between this protein level and the lowest arterial oxygen concentration, suggesting its potential involvement in the pathogenesis of obstructive sleep apnea. Further research is needed to explore the potential role and value of this protein.

Detecting serum galactomannan to diagnose acute invasive Aspergillus sinusitis: a meta-analysis

Summary: The serum galactomannan test showed limited accuracy in diagnosing confirmed AIAS based on the meta-analysis. Results indicate that reliance solely on the serum galactomannan test for initial AIAS diagnosis may be insufficient, and further studies are needed for a more accurate assessment of its diagnostic value in AIAS patients.

Incidence of Olfactory Dysfunction and Associated Factors: A Nationwide Cohort Study From South Korea

Summary: This study aimed to evaluate the prevalence of clinically diagnosed olfactory dysfunction in South Korea and its associations with neurodegenerative disorders. The results showed that olfactory dysfunction is underdiagnosed in South Korea, but it is clinically important considering the possibility of being a presymptom of neurodegenerative disorders. The study suggests that in clinical practice, the association between olfactory dysfunction and neurodegenerative disorders and possibly other systemic conditions should be considered.

Association between diabetes mellitus and chronic rhinosinusitis with nasal polyps: A population-based cross-sectional study

Is obstructive sleep apnea associated with erythrocytosis? A systematic review and meta-analysis

Clinical Characteristics and Surgical Outcomes of Obstructive Sleep Apnea Patients With Mixed Apnea Components

Summary: This study found that the presence of mixed apnea (MA) events in patients with obstructive sleep apnea (OSA) leads to different clinical characteristics and surgical outcomes. The mixed group had a lower surgical success rate and smaller postoperative improvements in sleep apnea indices compared to the pure group. Logistic regression analysis confirmed that the presence of MA is an independent predictor of poor surgical outcomes in OSA patients.

Comparison of clinical features and surgical outcomes between hypopnea- and apnea-predominant obstructive sleep apnea

Summary: This study investigated the clinical features and surgical outcomes of patients with hypopnea- and apnea-predominant obstructive sleep apnea (OSA). The results showed that patients with apnea-predominant OSA had more severe disease but comparable surgical outcomes compared to those with hypopnea-predominant OSA. Additionally, multilevel upper airway surgery may induce the transition from apnea to hypopnea in patients with apnea-predominant OSA.

Changes in Subjective Outcomes during the Early Period after Septoturbinoplasty

Summary: This study aimed to investigate changes in subjective outcomes within 6 months after septoturbinoplasty. Retrospective analysis of medical records of patients who underwent septoturbinoplasty at Gangnam Severance Hospital was conducted. Significant decreases in SNOT-22 and obstruction scores were observed at 1 month after surgery, but no significant changes were found at 3 and 6 months after surgery. A larger difference between preoperative and 1-month postoperative SNOT-22 scores was significantly associated with an improvement in symptoms at 3 or 6 months after surgery.

IL-4 drastically decreases deuterosomal and multiciliated cells via alteration in progenitor cell differentiation

Summary: In this study, the effect of allergic inflammation on nasal epithelial cell transcriptomes at the single-cell level was investigated using scRNAseq. The results showed that IL-4 led to a decrease in multiciliated cells and loss of deuterosomal cells. Furthermore, cell-specific markers that may be important for studying respiratory inflammatory diseases were identified.

Clinical characteristics and associated factors of qualitative olfactory dysfunction

Summary: This study investigated the clinical characteristics and associated factors of parosmia and phantosmia in patients with subjective smell disturbance. The results showed that younger age and female sex were related to both parosmia and phantosmia. Patients with COVID-19 had younger ages and higher TDI scores. Patients with parosmia or phantosmia had higher TDI scores and experienced more disruption in daily life. Younger age and higher TDI score were identified as independent factors associated with both parosmia and phantosmia.
